Daun kelor (Moringa oleifera) merupakan tanaman tropis yang dikenal luas sebagai Miracle Tree atau “pohon kehidupan” karena memiliki kandungan nutrisi dan senyawa bioaktif yang tinggi. Dua parameter penting yang menentukan kualitas bubuk daun kelor adalah aktivitas antioksidan dan Total Phenolic Content (TPC), yang mencerminkan potensi fungsional tanaman sebagai sumber antioksidan alami. Analisis kedua parameter tersebut umumnya dilakukan menggunakan metode DPPH untuk aktivitas antioksidan dan Folin–Ciocalteu untuk TPC. Meskipun akurat, kedua metode tersebut memerlukan waktu analisis yang relatif lama (4–6 jam), penggunaan reagen kimia dalam jumlah besar, serta menghasilkan limbah berbahaya. Oleh karena itu, diperlukan metode analisis yang lebih cepat, efisien, dan ramah lingkungan. Spektroskopi Fourier Transform Infrared (FTIR) merupakan salah satu teknologi yang berpotensi digunakan sebagai metode analisis non-destruktif karena mampu memberikan hasil yang cepat tanpa preparasi sampel yang kompleks dan tanpa menghasilkan limbah kimia.
Penelitian ini bertujuan untuk mengkaji kemampuan teknologi FTIR yang dikombinasikan dengan metode kemometrik Partial Least Squares (PLS) dalam memprediksi kandungan aktivitas antioksidan dan TPC bubuk daun kelor secara cepat dan akurat, serta menentukan metode pretreatment spektrum yang paling optimal. Spektrum FTIR diperoleh menggunakan Spectrum Two FT-IR Spectrometer pada rentang bilangan gelombang 4000–400 cm⁻¹. Model kalibrasi dikembangkan dengan metode PLS dan dievaluasi berdasarkan nilai koefisien korelasi (r), koefisien determinasi (R²), Root Mean Square Error of Calibration (RMSEC), dan Residual Predictive Deviation (RPD).
Hasil penelitian menunjukkan bahwa teknologi FTIR yang dikombinasikan dengan metode PLS mampu memprediksi kandungan TPC dan aktivitas antioksidan bubuk daun kelor dengan tingkat akurasi yang sangat baik. Nilai TPC sampel berkisar antara 27,03–31,61 mg GAE/g, sedangkan aktivitas antioksidan (IC₅₀ DPPH) berkisar antara 10,40–20,34 μg/mL, yang mengindikasikan seluruh sampel memiliki aktivitas antioksidan sangat kuat. Pretreatment First Derivative memberikan performa terbaik untuk prediksi TPC dengan nilai r sebesar 0,9969, R² sebesar 0,9938, RMSEC sebesar 0,0904 mg GAE/g, dan RPD sebesar 13,31. Pada prediksi aktivitas antioksidan, pretreatment yang sama juga menghasilkan performa terbaik dengan nilai r sebesar 0,9995, R² sebesar 0,9990, RMSEC sebesar 0,1016 μg/mL, dan RPD sebesar 33,88, yang menunjukkan kategori excellent prediction accuracy. Model yang dikembangkan menghasilkan nilai prediksi yang sangat mendekati nilai aktual hasil analisis laboratorium, Model FTIR-PLS menunjukkan akurasi prediksi yang baik, dengan selisih rata-rata 0,065 mg GAE/g untuk TPC dan 0,09 µg/mL untuk IC50. Prediksi paling akurat diperoleh pada sampel SP_5_KTP untuk parameter TPC (selisih 0,01 mg GAE/g), serta sampel SP_6_KBJ dan SP_10_LAMTIM untuk parameter IC50 (selisih 0,02 µg/mL), menunjukkan bahwa model mampu memprediksi nilai aktual dengan sangat mendekati pada sampel-sampel tersebut. sehingga membuktikan bahwa metode FTIR-PLS, khususnya dengan pretreatment First Derivative.

Moringa oleifera is a tropical plant widely recognized as the "Miracle Tree" due to its exceptionally high nutritional value and rich content of bioactive compounds. Two important parameters used to evaluate the quality of Moringa leaf powder are antioxidant activity and Total Phenolic Content (TPC), both of which reflect its potential as a natural source of antioxidants. These parameters are commonly determined using the DPPH assay for antioxidant activity and the Folin–Ciocalteu method for TPC analysis. Although these conventional methods provide accurate results, they require relatively long analysis times (4–6 hours), consume considerable amounts of chemical reagents, and generate hazardous chemical waste. Therefore, there is a growing need for a rapid, efficient, and environmentally friendly analytical method. Fourier Transform Infrared (FTIR) spectroscopy has emerged as a promising non-destructive analytical technique because it provides rapid measurements with minimal sample preparation and without producing chemical waste. This study aimed to evaluate the capability of FTIR spectroscopy combined with Partial Least Squares (PLS) chemometric analysis to rapidly and accurately predict the antioxidant activity and Total Phenolic Content (TPC) of Moringa leaf powder, as well as to determine the most effective spectral pretreatment method. FTIR spectra were collected using a Spectrum Two FT-IR Spectrometer over the wavenumber range of 4000–400 cm⁻¹. Calibration models were developed using the PLS method and evaluated based on the correlation coefficient (r), coefficient of determination (R²), Root Mean Square Error of Calibration (RMSEC), and Residual Predictive Deviation (RPD). The results demonstrated that FTIR spectroscopy combined with PLS analysis was capable of accurately predicting both TPC and antioxidant activity in Moringa leaf powder. The TPC values ranged from 27.03 to 31.61 mg GAE/g, while the antioxidant activity (DPPH IC₅₀) ranged from 10.40 to 20.34 μg/mL, indicating that all samples possessed very strong antioxidant activity. The First Derivative pretreatment provided the best performance for TPC prediction, yielding a correlation coefficient (r) of 0.9969, a coefficient of determination (R²) of 0.9938, an RMSEC of 0.0904 mg GAE/g, and an RPD of 13.31. For antioxidant activity prediction, the same pretreatment also produced the best performance, with an r value of 0.9995, an R² of 0.9990, an RMSEC of 0.1016 μg/mL, and an RPD of 33.88, indicating excellent prediction accuracy. The developed FTIR–PLS model generated predicted values that closely matched the laboratory reference values, with average prediction errors of only 0.065 mg GAE/g for TPC and 0.09 μg/mL for IC₅₀. The highest prediction accuracy for TPC was obtained for sample SP_5_KTP, with a difference of only 0.01 mg GAE/g between the predicted and measured values. For antioxidant activity, the most accurate predictions were achieved for samples SP_6_KBJ and SP_10_LAMTIM, each with a difference of only 0.02 μg/mL. These findings demonstrate that the FTIR–PLS approach, particularly when combined with the First Derivative spectral pretreatment, provides a rapid, accurate, non-destructive, and environmentally friendly alternative for predicting the antioxidant activity and Total Phenolic Content of Moringa leaf powder.
