Keamanan pangan pada komoditas daging perlu mendapat perhatian serius karena tingginya
risiko kontaminasi mikroba di pasar tradisional, termasuk oleh Salmonella sp. yang kini banyak
menunjukkan resistensi terhadap antibiotik akibat penggunaan yang tidak terkontrol. Penelitian ini
bertujuan menguji aktivitas antibakteri Infusa daun si khoh-khoh sebagai alternatif antimikroba
alami terhadap isolat Salmonella sp. resisten antibiotik yang diisolasi dari daging sapi di Pasar
Tradisional Banda Aceh. Tiga isolat resisten (U1(2), U2(2), L1(2)) dan satu isolat Salmonella
ATCC 14028 digunakan dalam uji aktivitas dengan metode difusi cakram. Infusa diperoleh
melalui perebusan daun dengan perbandingan 1:1 yang menghasilkan 190 ml ekstrak. Hasil
penelitian menunjukkan bahwa Infusa daun si khoh-khoh memiliki aktivitas antibakteri kategori
lemah, ditandai dengan terbentuknya zona hambat mulai dari konsentrasi 25% (1,4–2,05 mm) dan
peningkatan diameter zona hambat pada konsentrasi lebih tinggi yaitu 100%, dengan nilai terbesar
2,53 mm pada isolat U2(2). Namun, isolat L1(2) tidak menunjukkan respon hambatan pada semua
konsentrasi. Aktivitas antibakteri yang rendah diduga terkait struktur dinding sel bakteri Gram
negatif yang lebih kompleks serta penurunan senyawa volatile akibat proses pemanasan. Secara
keseluruhan, Infusa daun si khoh-khoh tidak berpotensi sebagai agen antibakteri alami terhadap
beberapa isolat Salmonella sp. resisten antibiotik.

Food safety in meat commodities requires serious attention due to the high risk of microbial contamination in traditional markets, including contamination by Salmonella sp., which has increasingly shown resistance to antibiotics as a result of uncontrolled use. This study aimed to evaluate the antibacterial activity of si khoh-khoh leaf infusion as an alternative natural antimicrobial against antibiotic-resistant Salmonella sp. isolates isolated from beef in Banda Aceh Traditional Markets. Three resistant isolates (U1(2), U2(2), L1(2)) and one Salmonella ATCC 14028 isolate were used in the activity test employing the disc diffusion method. The infusion was obtained by boiling the leaves at a ratio of 1:1, producing 190 mL of extract. The results showed that the si khoh-khoh leaf infusion exhibited weak antibacterial activity, as indicated by the formation of inhibition zones starting at a concentration of 25% (1.4–2.05 mm) and an increase in inhibition zone diameter at higher concentrations, namely 100%, with the largest value of 2.53 mm observed in isolate U2(2). However, isolate L1(2) showed no inhibitory response at all concentrations. The low antibacterial activity is presumed to be related to the more complex cell wall structure of Gram-negative bacteria as well as the reduction of volatile compounds due to the heating process. Overall, the si khoh-khoh leaf infusion does not have potential as a natural antibacterial agent against several antibiotic-resistant Salmonella sp. isolates.
