 <note>Pliek U is one of fermented coconut products. During the coconut fermentation process, the microorganisms involved vary widely. This research was conducted to characrerize morphology (the shape of colony, Gram staining, catalase test, oxidase test) and physiology (resistance test to pH 2, 3 and 4 and resistance to temperature 23oC, 37oC and 45oC) of five isolates (1, 23, 30, 49 and 58) growing on MRS agar at facultative anaerobic condition. The selection of these isolates was carried out based on the ability to grow on MRS agar which were incubated at 37oC for 48 hours.   The results of the morphological characterization test showed that isolates 1 and 49 were expected to be Leuconostoc sp. Isolates 23, 30 and 58 are Gram-negative coccus, catalase positive and oxidase negative bacteria. The results of the physiological characteristics test showed that the five isolates could grow at all tested pH and temperature. Identification of the five isolates was also carried out by molecular method (16S rRNA-PCR). However, optimization of the DNA extraction stage is needed in further research. &#13;
