ABSTRAK

Telah dilakukan penelitian formulasi kapsul ekstrak etanol umbi ubi kelapa ungu
(Dioscorea alata L.) sebagai antidiabetes. Ekstrak etanol umbi ubi kelapa ungu diperoleh
menggunakan metode maserasi menggunakan pelarut etanol 96%. Hasil karakterisasi
menunjukkan ekstrak etanol umbi ubi kelapa ungu memiliki kadar air 21,6%±0,29, kadar
abu total 9,21%±0,114, kadar sari larut air 44,16%±1,44 dan kadar sari larut etanol
67,5%±2,5. Hasil pengujian fitokimia menunjukkan bahwa ekstrak etanol umbi ubi
kelapa ungu mengandung senyawa alkaloid, flavonoid, saponin, tanin dan steroid.
Komposisi kapsul yang dibuat terdiri dari ekstrak etanol umbi ubi kelapa ungu 46,6%,
vivapur 101 46,6%, aerosil 3%, magnesium stearat 1% dan talkum 2%. Hasil evaluasi
massa kapsul memenuhi persyaratan yaitu sudut diam 16,12o ± 1,50, laju alir 7,13 g/s ±
0,88, Rasio Hauster’s dan indeks Carr’s 1,2 dan 6,32%. Hasil evaluasi sediaan
menunjukkan bahwa kapsul memiliki bobot yang seragam, waktu hancur yang memenuhi
persyaratan yaitu 2 menit 29 detik ± 12 detik dan kadar air isi kapusl yang baik yaitu
kurang dari 10%. Uji aktivitas antidiabetes dilakukan pada mencit (Mus muscullus)
dengan induktor diabetes berupa aloksan. Mencit dibagi kedalam 7 kelompok, dimana
kelompok 1 (kontrol negatif) diberikan CMC-Na 0,5%, kelompok 2 (kontrol positif)
diberikan tablet Metformin. Kelompok 3 diberikan ekstrak etanol umbi ubi kelapa ungu
dengan dosis 1120 mg/kgBB manusia. Kelompok 4,5,6 dan 7 diberikan sediaan kapsul
dengan dosis 350 mg/kgBB manusia (1 kapsul), 700 mg/kgBB manusia (2 kapsul), 1050
mg/kgBB manusia (3 kapsul) dan 1400 mg/kgBB manusia (4 kapsul). Persentase
penurunan kadar gula darah pada hari ke-21 pemberian perlakuan yang diperoleh
berturut-turut 11,9%, 30,30%, 24,85%, 17,86%, 20,14%, 22,9% dan 34,82% dengan pvalue (sig.) < ? (0.05).

ABSTRACT

The ethanol extract of purple yam tuber (Dioscorea alata L.) capsules as an antidiabetic
has been conducted. Ethanol extract of purple yam tubers was obtained using maceration
method using 96% ethanol solvent. Characterization results showed ethanol extract of
purple yam tubers had a water content of 21.6% ± 0.29, total ash content of 9.21% ±
0.114, water soluble extract content of 44.16% ± 1.44 and ethanol soluble extract content
of 67, 5% ± 2.5. Phytochemical screening results showed that the ethanol extract of
purple yam tubers contained alkaloid compounds, flavonoid, saponins, tannins and
steroids. The composition of the capsules consisted of ethanol extract of 46.6% purple
coconut sweet potato, 46.6% vivapur 101, 3% aerosil, 1% magnesium stearate and 2%
talc. The results of the capsule mass evaluation meet the requirements of a standpoint of
16.12 ± 1.50, a flow rate of 7.13 g / s ± 0.88, a Hauster's ratio and a Carr's index of 1.2
and 6.32%. The results of the preparation of the preparations showed that the capsules
had uniform weights, disintegration times that met the requirements of 2 minutes 29
seconds ± 12 seconds and the requirements of water content is less than 10%. Antidiabetic
activity test was carried out on mice (Mus muscullus) with diabetes inductor in the form
of alloxan. Mice were divided into 7 groups, group 1 was negative control in the form of
CMC-Na 0.5%, group 2 was positive control in the form of Metformin tablets. Group 3
is in the form of ethanol extract of purple coconut tubers at a dose of 1200 mg / kg human.
Groups 4,5,6 and 7 are capsules at a dose of 350 mg / kg human (1 capsule), 700 mg / kg
human (2 capsules), 1050 mg / kg human (3 capsules) and 1400 mg / kg human (4
capsule). The percentage decrease in blood sugar levels on the 21st day of treatment
obtained were 11.9%, 30.30%, 24.85%, 17.86%, 20.14%, 22.9% and 34.82, respectively.
