Metabolit sekunder daun pala diisolasi dengan maserasi 3500 g serbuk daun pala &#13;
menggunakan pelarut metanol dan diperoleh ekstrak sebanyak 535,7 g (15,3%) dengan&#13;
nilai IC&#13;
50 &#13;
188,4 ppm, dilanjutkan partisi dengan pelarut n-heksana, etil asetat dan&#13;
kloroform, diperoleh ekstrak masing-masing 1,8488 g (0,35%), 0,501g (0,15%) dan&#13;
7,122 g (2,28%) dengan nilai IC&#13;
50 &#13;
96,67 ppm, 0,216 ppm dan 35,62 ppm. Ekstrak nheksana&#13;
&#13;
dan kloroform diisolasi lebih lanjut dengan kromatografi kolom gravitasi&#13;
menggunakan eluen masing-masing n-heksana : etil asetat (9:1) dan kloroform : etil&#13;
asetat (8:2) dengan hasil diperoleh masing-masing 7 fraksi gabungan (MFHD 1-7) dan 6&#13;
fraksi gabungan (MFKD 1-6) dimana seluruh fraksi gabungan aktif sebagai antioksidan.&#13;
Fraksi gabungan dengan aktivitas tertinggi yaitu MFHD 2 (IC&#13;
50 &#13;
96,716 ppm), MFHD 4&#13;
(IC&#13;
50 &#13;
0,729 ppm), MFKD 4 (IC&#13;
50 &#13;
69,132 ppm) dan MFKD 5 (IC&#13;

direkromatografi dan dimurnikan dengan KLT preparatif, diperoleh fraksi dengan pola&#13;
noda tunggal pada kromatogram KLT yaitu  MFHD 2.3 (5,4 mg), MFHD 4.4 (10,7 mg)&#13;
dan MFKD 4.4 (5,8 mg) dengan IC&#13;
50 &#13;
masing-masing  42,299 ppm, 44,084 ppm dan&#13;
111,91 ppm. Hasil analisis GC-MS fraksi MFHD 2.3 mengandung senyawa eucalyptol&#13;
dan sitostenone. Berdasarkan beberapa referensi kedua senyawa tersebut memiliki&#13;
aktivitas antioksidan.&#13;

ABSTRACT&#13;
Isolation of secondary metabolites nutmeg leaf maceration begins with 3500 g nutmeg &#13;
leaf powder using methanol and obtained extract as much as 535.7 g (15.3%) with IC&#13;

value of 188.4 ppm, followed partition with n-hexane, ethyl acetate and chloroform,&#13;
each extract was obtained 1.8488 g (0.35%), 0,501g (0.15%) and 7.122 g (2.28%) with&#13;
IC&#13;
50 &#13;
value of 96.67 ppm, 0.216 ppm and 35.62 ppm. The extract n-hexane and&#13;
chloroform were isolated further by column chromatography gravity using eluent each&#13;
n-hexane : ethyl acetate (9 : 1) and chloroform : ethyl acetate (8 : 2) with the results&#13;
obtained by each of the 7 fractions combined (MFHD 1-7) and 6 fractions combined&#13;
(MFKD 1-6) in which all of the combined fractions are active as antioxidants. The&#13;
combined fraction with the highest activity MFHD 2 (IC50 96.716 ppm), MFHD 4&#13;
(IC50 0,729 ppm), MFKD 4 (IC50 69.132 ppm) and MFKD 5 (IC50 80.44 ppm)  return&#13;
column chromatography gravity and purified by preparative TLC, the fraction with a&#13;
pattern single spot on TLC chromatograms are MFHD 2.3 (5.4 mg), MFHD 4.4 (10.7&#13;
mg) and MFKD 4.4 (5.8 mg) with IC&#13;
50&#13;
 respectively 42.299 ppm, 44.084 ppm and&#13;
111.91 ppm. The GC-MS analysis of MFHD 2.3 fraction contains eucalyptol and&#13;
sitostenone compounds. Based on some references both of these compounds have&#13;
antioxidant activity. &#13;
