NON-INVASIVE STRESS HORMONE ANALYSIS IN SUMATRAN ELEPHANT (Elephas maximus

ABSTRACT

Non-invasive measurement of stress hormones is more effective to be applied to wildlife such as the Sumatran elephant. This study aims to measure the concentration of cortisol hormone metabolites in a non-invasive manner and its correlation with environmental factors, namely temperature and rainfall on a Sumatran elephant at the Elephant Conservation Center Saree, Aceh Besar. Samples were collected from five Sumatran elephants for five weeks (1 sample / head / week). Environmental temperature data, rainfall, and rectal temperature (body) were measured together with the collection of faecal samples. Fecal samples were extracted and the concentration of cortisol metabolites was measured using the enzyme-linked immunosorbent assay (ELISA) method. The data obtained were analyzed using a correlation test to determine the relationship between the concentration of cortisol metabolites and parameters of environmental temperature, rainfall, and rectal temperature (body). The mean (± SD) of cortisol metabolites concentration in Sumatran elephant faecal samples in weeks 1-5 were 138.46 ± 24.26 ng/g, 79.57 ± 14.91 ng/g, 98,. 21 ± 9.53 ng/g, 133.31 ± 12.42 ng/g and 108.67 ± 25.82 ng/g. Respectively the results of statistical analysis showed that increase of cortisol metabolites concentration was significant related with the increase of actual environmental temperature and body temperature (P?0.05). In conclusion, the concentration of cortisol metabolites in the fecal of Sumatran elephants has fluctuated and correlated with the actual environmental temperature and body temperature.
