KRUENG BARONA JAYA ACEH BESAR&#13;

ABSTRAK&#13;
Umumnya Escherichia coli bersifat non-patogen, namun ada beberapa &#13;
yang bersifat patogen dan membawa gen virulen. Penularan penyakit oleh&#13;
Escherichia coli dapat terjadi akibat mengkonsumsi makanan yang berasal dari&#13;
hewan ataupun produk olahannya, yang dikenal dengan istilah food borne disease.&#13;
Penelitian ini bertujuan untuk mengisolasi Escherichia coli patogen dari sampel&#13;
sapi diare di pasar hewan Cot Iri Kecamatan Krueng Barona Jaya, Aceh Besar.&#13;
Penelitian ini dilakukan dengan cara mengambil swab rektal pada sapi dengan&#13;
gejala diare lalu diinokulasikan ke media nutrient broth (NB), kemudian ditanam&#13;
pada media Eosin Methylene Blue Agar (EMBA) dan diamati secara makroskopis&#13;
morfologi koloninya, dilanjutkan dengan pewarnaan Gram dan diamati secara&#13;
mikroskopis. Selanjutnya dilakukan isolasi Escherichia coli patogen dari koloni&#13;
yang tumbuh dari EMBA ke media selektif Sorbitol Mac Conkey Agar (SMAC).&#13;
Secara makroskopis koloni Escherichia coli yang tumbuh pada media EMBA&#13;
tampak berwarna hijau metalik. Setelah pewarnaan Gram koloni bakteri terlihat&#13;
berwarna merah muda dan berbentuk batang pendek (kokobasil). Pada  media&#13;
SMAC koloni terlihat berwarna pucat (colourless) jika diamati secara makroskopis.&#13;
Hasil penelitian ini menunjukkan bahwa 6 dari 15 sampel feses sapi yang diteliti&#13;
positif terdapat Escherichia coli patogen. Oleh sebab itu disimpulkan bahwa 6 dari&#13;
15 sampel sapi diare  Escherichia coli patogen dapat diisolasi pada feses sapi diare&#13;
di pasar hewan Cot Iri Kecamatan Krueng Barona Jaya, Aceh Besar. &#13;

AT COT IRI ANIMAL MARKET OF THE DISTRICT &#13;
KRUENG BARONA JAYA ACEH BESAR&#13;

ABSTRACT&#13;
In most cases Escherichia coli are non-pathogenic, but some of them are &#13;
pathogenic and virulent carried. Transmission of Escherichia coli can occur because&#13;
of consume meat or the processed products, which is called foodborne disease. The&#13;
aim of this study to isolate pathogenic Escherichia coli  from diarrhea cattles&#13;
samples at Cot Iri animal market of the District Krueng Barona Jaya, Aceh Besar.&#13;
This research was conducted by taking rectal swabs in cattle and inoculated into&#13;
nutrient broth (NB) media, then planted on Eosin Methylene Blue Agar (EMBA)&#13;
media and the morphology of the colonie, observed macroscopically, followed by&#13;
Gram staining and observed microscopically. Furthermore, pathogenic Escherichia&#13;
coli colonies EMBA grown isolated to selective media Sorbitol Mac Conkey Agar&#13;
(SMAC). Macroscopically the colonies that grow on colonies EMBA media appear&#13;
metallic green sheen. After Gram staining, bacteria looks pink and have coccobacil&#13;
shape. The colonies that grow in SMAC appear to be pale color (colorless). The&#13;
conclution, indicate that 6 samples from this research were positively have&#13;
Escherichia coli pathogen. There were pathogenic Escherichia coli bacteria can be&#13;
isolated from 6 out of 15 samples faeces of diarrhea cattles at Cot Iri animal market&#13;
district Krueng Barona Jaya, Aceh Besar.  &#13;
