 <note>ABSTRAK&#13;
Penelitian ini bertujuan mempelajari kemampuan dari bakteri termo-halofilik isolat PLS 76 dalam menghasilkan antibiotik, mengidentifikasi spesies bakteri secara fenotip dan genotip. Aktivitas antibiotik bakteri termo-halofilik isolat PLS 76 diuji dengan metode difusi Kirby-Bauer. Identifikasi bakteri dilakukan secara fenotip  dengan pewarnaan Gram, SEM dan  uji biokimia and uji genotip dilakukan  melalui analisis gen 16S rRNA dengan metode Polymerase Chain Reaction (PCR). Hasil fermentasi bakteri termo-halofilik isolat PLS 76 pada media Tripton Soy Broth (TSB) 3,5% diketahui dapat menghasilkan antibiotik dengan diameter zona hambat sebesar 8 mm terhadap bakteri  uji E.coli dan tidak ada aktivitas zona hambat terhadap  S.aureus. Berdasarkan uji golongan antibiotik mengkonfirmasi bahwa isolat PLS 76 dikelompokkan pada golongan polipeptida. Identifikasi spesies bakteri secara fenotip menunjukkan bahwa bakteri termo-halofilik isolat PLS 76 merupakan bakteri Gram negatif, berbentuk batang (Basil) dan memiliki kemiripan secara biokimia dengan Bacillus lentus dan Bacillus megaterium. Identifikasi secara genotip memiliki kemiripan terdekat dengan tiga kelompok bakteri, yaitu uncultured organism, Bacillus dan Geobacillus dengan tingkat kemiripan sebesar 92%. Analisis pohon filogenetik untuk bakteri termo-halofilik isolat PLS 76 tidak memiliki kekerabatan terdekat dengan membuat kelompok grup tersendiri. Sehingga dapat disimpulkan bahwa bakteri termo-halofilik isolat PLS 76 dapat menghasilkan antibiotik golongan polipeptida dan merupakan genus Geobacillus  strain baru khas isolat lokal Pria Laot Sabang.&#13;

ABSTRACT&#13;
The aim of this research is to study the ability of thermo-halophilic bacterium of PLS 76 isolate in producing antibiotics, bacterial species identification is carried out by phenotype and genotype. Antibiotic activity of thermo-halophilic bacteria isolate PLS 76 was evaluated by Kirby-Bauer method. The bacterial identification was phenotyped by Gram staining, SEM and biochemical test and genotypes by analysis of 16S rRNA gene by Polymerase Chain Reaction (PCR) method. The result of fermentation of thermo-halophilic bacteria isolate PLS 76 on 3.5% of Tripton Soy Broth (TSB) identified be able to produce antibiotic with 8 mm inhibition zone diameter against E.coli test bacteria and no inhibitory zone activity to S.aureus. Based on the class antibiotic test isolates PLS 76 grouped on the polypeptide group. Phenotypic identification of bacterial species showed that PLS 76 thermophilic bacteria 76 is a Gram-negative, rod-shaped (Bacillus) and biochemically similar to Bacillus lentus and Bacillus megaterium, while genotypically having the closest similarity to three groups of bacteria, the Uncultured organism , Bacillus and Geobacillus with a similarity rate of 92%. Phylogenetic tree analysis for thermophilic bacteria thermophilic isolates PLS 76 did not have the closest kinship to make a group of its own. So it can be concluded that the thermo-halophilic bacterium of PLS 76 isolate can produce polypeptide class antibiotics and is a new genus of Geobacillus a typical local strain of Pria Laot Sabang isolate.&#13;
