EFEK PERBEDAAN WAKTU PENGAMBILAN SAMPEL DARAH DAN FESES TERHADAP KONSENTRASI HORMON KORTISOL PADA SAPI ACEH ABSTRAK Salah satu parameter yang harus dievaluasi sebelum dilakukan analisis hormon kortisol adalah waktu koleksi sampel. Hal ini karena, terdapat variasi diurnal konsentrasi kortisol pada pagi dan siang hari. Oleh karena itu, penelitian ini bertujuan untuk mengukur konsentrasi kortisol di dalam darah dan feses yang dikoleksi pada pagi dan siang hari untuk mengevaluasi ada tidaknya variasi diurnal pada sapi aceh. Sampel yang digunakan dalam penelitian ini adalah 16 sampel darah dan feses (8 sampel di pagi hari dan 8 sampel di siang hari) yang dikoleksi dari 8 sapi aceh betina umur 2-3 tahun. Sampel darah dipreparasi menjadi plasma, sedangkan sampel feses diekstrak selanjutnya dilakukan pengukuran hormon kortisol menggunakan teknik enzymelinked immunosorbent assay (ELISA). Data yang diperoleh diuji menggunakan uji-t. Hasil penelitian menunjukkan konsentrasi hormon kortisol di dalam plasma pada pagi hari secara signifikan lebih tinggi 79,34% dibandingkan siang hari (P0,05). Kesimpulan, terdapat variasi diurnal hormon kortisol pada plasma sapi aceh pada pagi dan siang hari, tetapi pada sampel feses tidak terlihat variasi diurnal. xi The Effect of Different Time of Sample Collection of Blood and Feces on the Cortisol Concentration in Aceh Cattle ABSTRACT One of the parameter that must be evaluated prior to perform the cortisol hormone analysis is time of sample collection. This is due to there is diurnal variation of cortisol concentrations in the morning and noon. Therefore,this study aims to measure cortisol concentration in the blood and feces collected in the morning and noon for evaluating the diurnal variation in aceh cattle. Samples used in this study were 16 blood and feces samples (8 samples in the morning and 8 samples in the noon) collected from eight adult females of aceh cattle age 2-3 years old. Blood samples were prepared into a plasma, while the feces samples were extracted and then performed cortisol hormone measurement through enzyme-linked immunosorbent assay (ELISA) technique. Data were analyzed by using t-test. The result showed that concentrations of cortisol in the plasma collected in the morning were significantly higher 79,34% compared to the noon samples (p 0,05).
