Penelitian ini bertujuan untuk menguji stabilitas dan menentukan struktur antibiotik golongan ?-Laktam dari bakteri termohalofilik isolat PLS 80. Uji stabilitas antibiotik dilakukan dengan melakukan penyimpanan antibiotik pada variasi waktu dan uji aktivitasnya dilakukan dengan metode Kirby-Bauer, sementara struktur antibiotik diidentifikasi menggunakan instrumen Liquid Chromatoghrapy-Mass Spectrometry (LC-MS). Uji aktivitas ekstrak kasar antibiotik menunjukkan bahwa fraksi metanol pekat dari bakteri termohalofilik isolat PLS 80 menghasilkan zona hambat sebesar 11 mm terhadap E. coli dan 12 mm terhadap S. aureus. Antibiotik dari bakteri termohalofilik isolat PLS 80 dimurnikan menggunakan Kromatografi Lapis Tipis Preparatif (KLTP). Hasil uji stabilitas antibiotik selama masa penyimpanan menunjukkan antibiotik lebih stabil pada waktu 24 jam dibandingkan waktu 2 dan 48 jam yang ditunjukkan dengan diameter zona hambat paling baik pada masa penyimpanan 24 jam terhadap E. coli dan S. aureus dengan masing-masing diameter zona hambat sebesar 8 mm. Struktur senyawa antibiotik yang diidentifikasi menggunakan LC-MS menunjukkan bahwa bakteri termohalofilik isolat PLS 80 diduga mampu menghasilkan antibiotik dari kelompok amoksisilin yang merupakan turunan antibiotik penisilin.&#13;

ABSTRACT&#13;
The aim of this researh is to evaluate the stability and to determine the structure of antibiotic from thermohalophilic bacterium of PLS 80 isolate. Stability test of the antibiotic was carried out by varying the antibiotic storage time. The activity was examined by Kirby-Bauer method, meanwhile the antibiotic structure was identified by using Liquid Chromatography-Mass Spectrometry (LC-MS). The activity test of crude extract of the antibiotic exhibited that the concentrated methanol fraction from PLS 80 isolate resulted inhibition zone of 11 mm against E. coli and 12 mm against S. aureus. The antibiotic from thermohalophilic bacterium of PLS 80 isolate was purified by using Preparative Thin Layer Chromatography (Prep TLC). The result of stability test showed that the antibiotic is more stable after 24 hours storage in comparison to 2 and 48 hours storage time. The structure of the antibiotic was identified by using LC-MS. PLS 80 isolate seemed to be able to produce amoxicillin antibiotic which is a penicillin derivative.&#13;
