Penelitian ini bertujuan untuk menentukan hasil reaksi transesterifikasi dengan&#13;
katalis lipase dari bakteri termo-halofilik isolat PLS 80 serta mengetahui kualitas&#13;

dan aerasi 150 rpm. Pemurnian lipase dilakukan menggunakan fraksinasi&#13;
aseton dimana diperoleh fraksi 25-44% merupakan fraksi yang memiliki nilai&#13;
aktivitas spesifik tertinggi yakni sebesar 16,863 unit/mg protein. Lipase yang telah&#13;
dimurnikan selanjutnya digunakan dalam reaksi transesterifikasi minyak kelapa&#13;
dan metanol (dengan perbandingan molar 6:1) menghasilkan metil ester. Analisis&#13;
GS-MS menunjukkan bahwa biodiesel yang diperoleh mengandung metil&#13;
oktanoat (5,3%), metil kaprat (12,41%), metil laurat (34,13%), metil miristat&#13;
(10,73%), metil palmitat (3,93%) dan metil stearat (1,20%).  Biodiesel yang&#13;
dihasilkan memiliki densitas sebesar 0,90 g/cm&#13;

 dan viskositas sebesar 9,27 cSt&#13;
dan nyala api kurang dari dua detik. Lipase yang dihasilkan dari bakteri termohalofilik&#13;
&#13;
isolat Pria Laot Sabang (PLS) 80 dapat mengkatalisis reaksi&#13;
transesterifikasi menghasilkan biodiesel.&#13;

ABSTRACT &#13;
The goals of this research are to determine transesterification reaction by lipase&#13;
catalyst from thermo-halophilic bacteria of PLS 80 isolate and to identify the&#13;
biodiesel quality. Thermo-stable lipase from PLS 80 isolate was produced in ½&#13;
thermus medium for 16 hours with 150 rpm agitation. The lipase purification was&#13;
carried out by aceton fractionation where fraction 25-44% gave the highest&#13;
specific activity value of 16.863 units/ mg protein. The purified lipase was used in&#13;
transesterification of coconut oil and methanol (which molar ratio 6:1). GC-MS&#13;
analyzed showed that the biodiesel is consisted of methyl octanoate (5.3%),&#13;
methyl caprate (12.41%), methyl laurate (34.13%), methyl myristate (10.73%),&#13;
methyl palmitate (3.93%) and methyl stearate (1,20%). The biodiesel has 0.90&#13;
g/cm&#13;

density; 9.27 cSt viscocity and flame less than 2 seconds. It can be&#13;
concluded that the obtained lipase from PLS 80 isolate was able to catalysed&#13;
transesterification to produce biodiesel. &#13;
