Penelitian ini bertujuan untuk memurnikan dan mengkarakterisasi lipase dari bakteri termo-halofilik isolat PLS 80. Lipase diproduksi pada media ½ thermus selama 16 jam, suhu 70oC dan aerasi 150 rpm. Pemurnian dilakukan menggunakan fraksinasi ammonium sulfat, dilanjutkan dengan kromatografi filtrasi gel sepharose Cl-6B. Berat molekul lipase murni adalah ±50 KDa setelah dianalisis menggunakan SDS-PAGE dan zymografi menggunakan substrat naftil asetat. Karakterisasi lipase menunjukkan aktivitas optimum pada suhu 70oC dan pH basa (pH 9). Penambahan 10 mM ion logam  Mn2+, K+, Ca2+, and Hg2+  diketahui dapat meningkatkan aktivitas lipase, sedangkan penambahan 10 mM ion logam Mg2+, Zn2+, Co2+ serta inhibitor EDTA, dan PMSF diketahui dapat menurunkan aktivitas lipase. Hasil yang diperoleh menyarankan bahwa lipase yang dihasilkan dari bakteri termo-halofilik isolat PLS 80 termasuk ke dalam jenis lipase termostabil, alkali, kelompok lipase serin dan metaloenzim. &#13;

ABSTRACT  &#13;
This study aimed to purify and characterize lipase from thermo-halophilic bacteria PLS 80. Lipase  was produced in  ½ thermus medium for 16 hours, 70oC and agitation at 150 rpm. Purification was performed using ammonium sulfate fractionation, followed by gel filtration chromatography using Sepharose Cl-6B. Molecular weight of the purified lipase was about ±50 KDa after SDS-PAGE analysis and zymography with substrat of alfa naftil acetate. The characterization of purified thermostable lipase showed an optimum activity at 70°C and alkaline pH  (pH 9). The addition of 10 mM metal ions Mn2+, K+, Ca2+, and Hg2+ increased lipase activity, while the addition of 10 mM Mg2+, Zn2+, Co2+, EDTA, and PMSF decreased lipase activity. The results suggested that the lipase that produced from thermo-halophilic bacteria PLS 80 belongs to the type of thermostable lipase, alkaline, serine lipase group and metalloenzyme. &#13;
