 <note>Most probable number and membrane filter were methods that can be used for&#13;
analyzing of  Escherichia coli contained in refill drinking water. Based on&#13;
Permenkes 492/Menkes/Per/IV/2010 about the Regulations of Drinking Water&#13;
Quality, the content of Escherichia coli in a 100 ml sample is zero. The research&#13;
aimed to know the differences in the content of Escherichia coli in refill drinking&#13;
water which were analized by Most probable number  method and Membrane&#13;
filter method. The research used observation method with descriptive analytic&#13;
design which is carried out on 28 samples of drinking water refill depot in city&#13;
center and downtown area. The data were analyzed by t-test and Anova using&#13;
SPSS version 16. The results showed that there is a significant difference between&#13;
the membrane filter method and most probable number method. Anova test&#13;
showed no significant difference between the downtown area and the city center&#13;
either to the monitored or unmonitored region. The average of Escherichia coli&#13;
content observed using membrane filter method was 4,46 CFU/100 ml sample&#13;
while the content with most probable number method was 2,64 MPN Escherichia&#13;
coli/100 ml sample. Therefore the membrane filter method was more sensitive&#13;
than most probable number method in analizing the content of Escherichia coli in&#13;
refill drinking water.&#13;
