Dielektroforesis (DEP) merupakan teknik manipulasi partikel yang banyak diterapkan pada platform mikrofluida berbasis mikroelektroda. Namun analisis kuantitatif pergerakan partikel pada fenomena dielektroforesis yang dilakukan secara manual masih rentan terhadap subjektivitas dan tidak efisien untuk data berjumlah besar. Penelitian ini bertujuan mengembangkan software pipeline berbasis Python untuk melacak dan menganalisis secara kuantitatif pergerakan biopartikel pada fenomena DEP positif (pDEP) dan DEP negatif (nDEP), serta mengkarakterisasi dinamika pergerakannya. Metode penelitian terdiri dari pengembangan algoritma pengolahan citra menggunakan OpenCV untuk deteksi partikel per frame, TrackPy untuk pelacakan multi-objek, serta ekspor data koordinat ke format CSV untuk analisis lanjutan berupa kecepatan, jarak terhadap titik tangkap biopartikel pada elektroda, dan tren pergerakan antar frame, dengan data rujukan hasil eksperimen D'Amico et al. (2017) pada mikroelektroda quadrupole yang menggunakan sampel biopartikel. Hasil penelitian menunjukkan bahwa pipeline yang dikembangkan mampu mendeteksi, melacak lintasan biopartikel pada masing-masing kondisi DEP secara akurat, dan mengkarakterisasi dinamika pergerakannya. Pada kondisi nDEP, biopartikel bergerak menjauhi elektroda menuju titik tangkap di tengah elektroda dengan kecepatan rata-rata 26,63 px/s, sedangkan pada kondisi pDEP biopartikel bergerak ke titik tangkap di tepi elektroda dengan kecepatan rata-rata lebih rendah yaitu 12,20 px/s. Perbedaan pola gerakan biopartikel ini dipengaruhi oleh sifat dielektrik biopartikel, frekuensi medan listrik, konduktivitas listrik medium dan biopartikel, tegangan listrik, medan listrik, dan geometri mikroelektroda yang menentukan arah dan besar gaya DEP. Penelitian ini bermanfaat sebagai prototipe software tool open-source berbasis Python untuk analisis data eksperimen bidang mikrofluida dan elektrokinetik, serta menjadi alat bantu edukatif dalam pengembangan kemampuan pemrosesan citra digital dan pemrograman Python.

Dielectrophoresis (DEP) is a particle manipulation technique widely applied in microelectrode-based microfluidic platforms. However, quantitative analysis of particle movement in dielectrophoresis phenomena performed manually remains prone to subjectivity and is inefficient for large datasets. This study aims to develop a Python-based software pipeline to track and quantitatively analyze the movement of bioparticles under positive DEP (pDEP) and negative DEP (nDEP) phenomena, as well as to characterize their movement dynamics. The research method consisted of developing image processing algorithms using OpenCV for particle detection per frame, TrackPy for multi-object tracking, and exporting coordinate data to CSV format for further analysis including velocity, distance relative to the bioparticle capture point on the electrode, and movement trends across frames. Experimental results from D'Amico et al. (2017) on quadrupole microelectrodes using bioparticle samples were used as reference data. The results showed that the developed pipeline was capable of accurately detecting and tracking bioparticle trajectories under each DEP condition, as well as characterising their movement dynamics. Under the nDEP condition, bioparticles moved away from the electrodes toward the capture point at the center of the electrode array with an average velocity of 26.63 px/s, whereas under the pDEP condition, bioparticles moved to the capture point at the edge of the electrode with a lower average velocity of 12.20 px/s. This difference in movement patterns was influenced by the dielectric properties of the bioparticles, electric field frequency, electrical conductivity of the medium and bioparticles, applied voltage, electric field strength, and microelectrode geometry, which collectively determined the direction and magnitude of the DEP force. This study is useful as an open-source Python-based software tool prototype for analyzing experimental data in the fields of microfluidics and electrokinetics, while also serving as an educational aid in developing digital image processing and Python programming skills.
