Kanker merupakan pembentukan sel-sel abnormal secara cepat yang tumbuh di luar batas normal dan kemudian dapat menyebar ke organ lainnya. Perkembangan kanker dapat diatasi dengan menghambat pembentukan pembuluh darah baru melalui pemberian terapi obat, Salah satunya penggunaan obat herbal dari daun kari (Murraya koenigii) yang terbukti bersifat antikanker dan antioksidan. Penelitian ini bertujuan untuk melihat ekspresi CD34 pascaterapi ektrak etanol daun kari terhadap kanker kulit tikus putih (Sprague dawley) dan menentukan dosis terbaik untuk menjadi pengobatan alternatif herbal. Penelitian ini menggunakan 12 ekor tikus Sprague dawley jantan yang dibagi menjadi 4 kelompok. Kelompok negatif kulit normal (P0) kelompok tikus model kanker kulit tanpa diberikan terapi (P1), tikus model kanker kulit yang diterapi dengan ekstrak etanol daun kari dosis 200 mg/kg BB (P2), dan 300 mg/kg BB (P3) selama 30 hari. Pada hari ke-31 tikus dieutanasia dan dikoleksi sampel organ kulit yang teridentifikasi kanker, kemudian dibuat preparat histopatologi yang diwarnai dengan pewarnaan imunohistokimia (IHK) menggunakan marker CD34. Pengamatan ekspresi CD34 dilakukan dengan 8 lapang pandang dan di skoring dengan kategori skor 1: Lemah (coklat muda), skor 2: Sedang (coklat) dan skor 3: Kuat (coklat gelap). Hasil pengamatan menunjukkan skor rata-rata ekspresi CD34 kelompok P0 (7,0±2,7), kelompok P1 (51,8±13,8), kelompok P2 (31,5±8,9), dan kelompok P3 (18,0±7,0). Berdasarkan hasil yang diperoleh dapat disimpulkan bahwa pemberian terapi dengan ekstrak etanol daun kari dengan dosis 300mg/kg BB menujukkan rerata ekspresi CD34 lebih rendah dibandingkan kelompok model kanker kulit dan dosis 200 mg/kg BB.

Cancer is characterized by the rapid proliferation of abnormal cells that grow beyond normal boundaries and can spread to other organs. Cancer progression may be inhibited by suppressing the formation of new blood vessels through therapeutic agents. One potential herbal therapy is curry leaf (Murraya koenigii), which has demonstrated anticancer and antioxidant properties. This study aimed to evaluate CD34 expression following treatment with ethanolic extract of curry leaves in a white rat (Sprague Dawley) skin cancer model and to determine the most effective dose as a potential herbal alternative therapy. This study used 12 male Sprague Dawley rats divided into four groups: normal skin (P0), skin cancer model without treatment (P1), skin cancer model treated with ethanolic curry leaf extract at a dose of 200 mg/kg body weight (P2), and 300 mg/kg body weight (P3) for 30 days. On day 31, the rats were euthanized, and skin tissues identified as cancerous were collected for histopathological examination. The tissue sections were stained using immunohistochemistry (IHC) with CD34 as the primary marker. CD34 expression was evaluated in eight microscopic fields and scored according to staining intensity as follows: score 1 = weak (light brown), score 2 = moderate (brown), and score 3 = strong (dark brown). The mean CD34 expression scores were 7,0±2,7 in P0, 51.8±13,8 in P1, 31.5±8,9 in P2, and 18,0±7,0 in P3. These findings indicate that treatment with ethanolic curry leaf extract at a dose of 300 mg/kg body weight resulted in a lower mean CD34 expression than both the untreated skin cancer group and the 200 mg/kg body weight treatment group.
