Cendawan Dark Septate Endophyte (DSE) merupakan kelompok cendawan endofit dengan hifa bersepta berpigmen gelap yang mengandung melanin dan mampu mengkolonisasi akar tanaman tanpa menimbulkan gejala penyakit. DSE memiliki potensi sebagai agens hayati karena dapat menghambat perkembangan patogen sekaligus mendukung pertumbuhan tanaman. Tanaman nilam Aceh (Pogostemon cablin Benth.), sebagai komoditas penghasil minyak atsiri bernilai ekonomi tinggi, masih menghadapi kendala produktivitas akibat serangan penyakit budok (Synchytrium pogostemonis) dan layu bakteri (Ralstonia solanacearum dan Enterobacter sp.). Oleh karena itu, pemanfaatan DSE sebagai pendekatan pengendalian penyakit yang ramah lingkungan menjadi relevan dan penting. Penelitian ini bertujuan untuk mengisolasi dan mengkarakterisasi cendawan DSE dari akar tanaman nilam Aceh yang sehat.
Penelitian dilaksanakan di Laboratorium Ilmu Penyakit Tumbuhan, Fakultas Pertanian Universitas Syiah Kuala pada Februari–Juni 2026. Sampel akar nilam sehat diambil dari dua lokasi berbeda, yaitu Desa Teuladan, Kecamatan Seulimum, Kabupaten Aceh Besar dan Desa Blang Jorong, Kecamatan Bandar, Kabupaten Bener Meriah. Isolasi dilakukan menggunakan media Corn Meal Agar (CMA), pemurnian pada Potato Dextrose Agar (PDA), dan karakterisasi melalui pengamatan morfologi makroskopis serta mikroskopis.
Hasil eksplorasi terhadap enam sampel akar nilam Aceh menghasilkan dua isolat DSE, masing-masing dari Aceh Besar dan Bener Meriah. Secara makroskopis, koloni menunjukkan variasi warna abu-abu kehitaman hingga hitam dengan tekstur menyerupai kapas. Secara mikroskopis, kedua isolat memperlihatkan ciri khas DSE berupa hifa bersepta berpigmen gelap dengan percabangan jelas. Isolat dari Bener Meriah menunjukkan struktur hifa membengkak (swollen hyphal cells), sedangkan isolat dari Aceh Besar memperlihatkan hifa melingkar (hyphal coils). Tidak ditemukan konidia maupun konidiofor pada kedua isolat.
Penelitian ini menegaskan keberadaan cendawan DSE pada akar tanaman nilam Aceh, yang sebelumnya belum banyak dilaporkan. Kontribusi utama penelitian ini adalah memberikan bukti awal tentang keanekaragaman mikroorganisme endofit pada nilam Aceh serta membuka peluang pemanfaatan DSE sebagai agens hayati dalam sistem pengendalian penyakit tanaman yang berkelanjutan. Dengan demikian, hasil penelitian ini dapat menjadi dasar bagi pengembangan strategi budidaya nilam berbasis mikroba endofit yang ramah lingkungan. Meskipun demikian, identifikasi molekuler tetap diperlukan untuk memastikan identitas kedua isolat hingga tingkat taksonomi yang lebih akurat, sehingga pemanfaatannya dapat diarahkan secara lebih tepat dalam program budidaya nilam berkelanjutan.

Dark Septate Endophyte (DSE) fungi are a group of endophytic fungi with darkly pigmented, septate hyphae containing melanin that can colonize plant roots without causing disease symptoms. DSE has potential as a biological agent because it can inhibit pathogen development while supporting plant growth. Aceh patchouli (Pogostemon cablin Benth.), a commodity producing high-value essential oils, still faces productivity constraints due to attacks by budok disease (Synchytrium pogostemonis) and bacterial wilt (Ralstonia solanacearum and Enterobacter sp.). Therefore, the use of DSE as an environmentally friendly disease control approach is relevant and important. This study aims to isolate and characterize DSE fungi from the roots of healthy Aceh patchouli plants. The research was conducted at the Plant Pathology Laboratory, Faculty of Agriculture, Syiah Kuala University, from February to June 2026. Healthy patchouli root samples were collected from two different locations: Teuladan Village, Seulimum District, Aceh Besar Regency, and Blang Jorong Village, Bandar District, Bener Meriah Regency. Isolation was performed using Corn Meal Agar (CMA), purification on Potato Dextrose Agar (PDA), and characterization through macroscopic and microscopic morphological observations. The exploration of six Acehnese patchouli root samples yielded two DSE isolates, one from Aceh Besar and one from Bener Meriah. Macroscopically, the colonies varied in color from blackish gray to black with a cotton-like texture. Microscopically, both isolates exhibited the typical DSE characteristics of darkly pigmented, septate hyphae with distinct branching. The isolate from Bener Meriah exhibited swollen hyphal cells, while the isolate from Aceh Besar exhibited coiled hyphae. No conidia or conidiophores were found in either isolate. This study confirms the presence of DSE fungi in the roots of Acehnese patchouli plants, a previously unreported finding. The primary contribution of this study is providing preliminary evidence of the diversity of endophytic microorganisms in Acehnese patchouli and opening up opportunities for the use of DSE as a biological agent in sustainable plant disease control systems. Thus, the results of this study can form the basis for the development of environmentally friendly endophytic microbial-based patchouli cultivation strategies. However, molecular identification is still needed to confirm the identity of both isolates to a more accurate taxonomic level, so that their utilization can be more appropriately directed in sustainable patchouli cultivation programs.
