Acne vulgaris merupakan salah satu penyakit kulit yang menyerang hampir pada semua orang, yang.dapat mengurangi kenyamanan pencerita akibat rasa nyeri apabila mengalami inflamasi. Propionibacterium acnes (dikenal juga sebagai Cutibacterium acnes) merupakan salah satu patogen oportunistik yang memicu respon inflamasi acne vulgaris. Penggunaan obat artifisial dalam terapi acne vulgaris dapat menimbulkan efek samping sehingga dibutuhkan alternatif dari bahan alam yang lebih aman. Penelitian ini dilakukan untuk menganalisis prediksi potensi senyawa aktif campuran empat rempah (jahe, kunyit, serai dan jintan hitam) sebagai agen antiinflamasi pada acne vulgaris yang diinduksi P. acnes melalui analisis molecular docking serta mempelajari aktivitas sinyal molekuler dan jalur biologis terkait inflamasi acne vulgaris yang dipengaruhi oleh senyawa aktif melalui analisis network pharmacology. Hasil penelitian menunjukkan terdapat 13 senyawa dari campuran empat rempah tersebut yang memiliki prediksi aktivitas antiinflamasi tinggi dengan nilai probability activity (Pa)≥0,7, yaitu beta-Terpineol, 2H-inden-2-one,1,4,5,6,7,7a-hexahydro-7a-methyl-,(S)-, 7-epi-cis-sesquisabinene hydrate, geranyl acetate, humulen-(v1), curcumene, humulenol-II, (E)-atlantone, ethyl 9.cis.,11.trans.-octadecadienoate, ethyl linolelaidate, (6)-shogaol, cis-11,14-eicosadienoic acid, methyl ester, dan dihydroxanthin. Hasil analisis network pharmacology diperoleh 27 gen/protein target penting yang terkait dengan senyawa-senyawa terpilih dan terlibat dalam mekanisme inflamasi acne vulgaris oleh P. acnes. Hasil simulasi docking dengan mempertimbangkan nilai binding affinity dan interaksi ligan senyawa dengan reseptor yang dibandingkan ligan kontrol, senyawa aktif yang memiliki prediksi pengikatan terbaik dengan reseptor JNK1 yang terlibat dalam jalur persinyalan inflamasi akibat patogenesis P. acnes adalah (6)-Shogaol dan Dihydroxanthin. Kedua ligan ini diduga dapat menghambat aktivitas protein JNK1 dengan baik, sehingga dapat menekan respon inflamasi acne vulgaris akibat induksi P. acnes.

Acne vulgaris is a skin condition that affects nearly everyone and can cause discomfort due to pain when inflammation occurs. Propionibacterium acnes (also known as Cutibacterium acnes) is an opportunistic pathogen that triggers the inflammatory response in acne vulgaris. The use of synthetic drugs in the treatment of acne vulgaris can cause side effects, making safer alternatives derived from natural sources necessary. This study was conducted to analyze the predicted potential of active compounds from a mixture of four spices (ginger, turmeric, lemongrass, and black cumin) as anti-inflammatory agents in P. acnes-induced acne vulgaris through molecular docking analysis, as well as to investigate the molecular signaling activities and biological pathways related to acne vulgaris inflammation that are influenced by these active compounds through network pharmacology analysis. The results showed that 13 compounds from the four-spice mixture were predicted to have high anti-inflammatory activity with a probability of activity (Pa) ≥ 0.7, namely beta-terpineol, 2H-inden-2-one, 1,4,5,6,7,7a-hexahydro-7a-methyl-, (S)-, 7-epi-cis -sesquisabinene hydrate, geranyl acetate, humulen-(v1), curcumene, humulenol-II, (E)-atlantone, ethyl 9-cis,11-trans-octadecadienoate, ethyl linolelaidate, (6)-shogaol, cis-11,14-eicosadienoic acid methyl ester, and dihydroxanthin. The network pharmacology analysis identified 27 key target genes/proteins associated with the selected compounds and involved in the inflammatory mechanism of acne vulgaris caused by P. acnes. The results of docking simulations which considered binding affinity and the interactions of the compounds’ ligands with receptors compared to control ligands, indicated that the active compounds with the best predicted binding to the JNK1 receptor, which is involved in the inflammatory signaling pathway resulting from P. acnes pathogenesis, are (6)-Shogaol and Dihydroxanthin. These two ligands are believed to effectively inhibit JNK1 protein activity, thereby suppressing the inflammatory response in acne vulgaris induced by P. acnes.
