Skin health is influenced by various factors, including the balance of the skin microbiota. One of the bacteria that plays a role in the skin ecosystem is Staphylococcus aureus, which under certain conditions may induce inflammation and impair the skin barrier. This study aimed to analyze the relationship between the quantity of Staphylococcus aureus and facial skin health among adult women in Banda Aceh City based on skin hyperpigmentation, hydration, and wrinkle parameters. This analytical observational study employed a cross-sectional design involving 48 women aged 30–40 years using stored biological samples in the form of facial skin swabs. The quantity of Staphylococcus aureus was measured using quantitative Polymerase Chain Reaction (qPCR) based on cycle threshold (CT) values, while skin health parameters were assessed using the Magic Mirror Plus® Skin Analyzer. Data were analyzed using univariate and bivariate analyses with correlation tests according to data distribution. The results showed that the mean CT value of Staphylococcus aureus was 35.34 ± 1.84, while the mean hyperpigmentation, hydration, and wrinkle levels were 25.75% ± 13.56, 63.19% ± 12.49, and 39.17% ± 16.42, respectively. There were no significant correlations between the quantity of Staphylococcus aureus and hyperpigmentation (r = 0.073; p = 0.621), hydration (r = –0.183; p = 0.213), or wrinkles (r = 0.057; p = 0.701). In conclusion, the quantity of Staphylococcus aureus was not significantly associated with facial skin health among adult women in Banda Aceh City.
