Spermatogenesis terjadi di dalam tubulus seminiferus yang mencakup spermatositogenesis dan spermiogenesis. Selama spermiogenesis, terjadi perkembangan akrosom yang terus meningkat sejak fase pubertas dengan melibatkan glikokonjugat N-asetilgalaktosamin. Penelitian ini bertujuan mengkaji tahapan perkembangan akrosom spermatozoa selama spermiogenesis serta distribusi N-asetilgalaktosamin pada testis kelinci lokal dengan umur berbeda menggunakan pewarnaan histokimia lektin peanut agglutinin (PNA). Penelitian ini menggunakan blok jaringan testis sinister dari 12 ekor kelinci lokal jantan yang dibagi menjadi empat kelompok umur, yaitu 2 bulan (K-I), 6 bulan (K-II), 12 bulan (K-III), dan 24 bulan (K-IV), masing-masing terdiri atas tiga ekor. Slide testis diwarnai menggunakan histokimia lektin PNA metode avidin-biotin complex (ABC). Tahapan perkembangan akrosom dan distribusi glikokonjugat dianalisis secara deskriptif, sedangkan perbedaan ekspresi dianalisis menggunakan uji Kruskal-Wallis dan Mann Whitney U. Hasil penelitian menunjukkan bahwa N-asetilgalaktosamin terdeteksi pada akrosom round spermatid, elongated spermatid, dan spermatozoa pada kelompok K-II, K-III, dan K-IV, tetapi tidak terdeteksi pada K-I. Perkembangan akrosom berlangsung dalam 14 steps yang tersebar pada 8 stages epitel tubulus seminiferus, dengan steps 1–4 berada pada fase Golgi, steps 5–7 fase cap, steps 8–11 fase akrosom, dan steps 12–14 fase maturasi. Analisis statistik ekspresi N-asetilgalaktosamin pada akrosom setiap stage menunjukkan adanya perbedaan nyata (P0,05) antar kelompok K-II, K-III, dan K-IV. Disimpulkan bahwa perkembangan akrosom kelinci berlangsung melalui 14 steps dalam 8 stages spermatogenesis, dengan ekspresi N-asetilgalaktosamin muncul setelah kelinci mencapai kematangan reproduksi.

Spermatogenesis occurs within the seminiferous tubules and comprises two phases: spermatocytogenesis and spermiogenesis. During spermiogenesis, the acrosome develops progressively from puberty onward, involving N-acetylgalactosamine-containing glycoconjugates. This study aimed to determine the stages of spermatozoa acrosomal development during spermiogenesis and to assess the distribution of N-acetylgalactosamine in the testes of local rabbit of different ages using peanut agglutinin (PNA) lectin histochemistry. Left testicular tissue blocks from 12 rabbits were used and divided into four age groups: 2 months (K-I), 6 months (K-II), 12 months (K-III), and 24 months (K-IV), with three rabbits in each group. Tissue sections were stained with PNA lectin histochemistry using the avidin-biotin complex (ABC) method. Acrosome developmental stages and glycoconjugate distribution were analyzed descriptively, while expression differences were evaluated using the Kruskal–Wallis test followed by the Mann–Whitney U test. The results showed that N-acetylgalactosamine was detected in the acrosomes of round spermatids, elongated spermatids, and spermatozoa in groups K-II, K-III, and K-IV, but was not detected in K-I. Acrosome development proceeded through 14 developmental steps distributed across eight stages of the seminiferous epithelium cycle. Steps 1–4 corresponded to the Golgi phase, steps 5–7 to the cap phase, steps 8–11 to the acrosome phase, and steps 12–14 to the maturation phase. Statistical analysis of N-acetylgalactosamine expression in the acrosome at each stage revealed significant differences (P < 0.05) between K-I and K-II, K-III, and K-IV, whereas no significant differences (P > 0.05) were observed among K-II, K-III, and K-IV. In conclusion, acrosome development in rabbit occurs through 14 distributed in 8 stages of spermatogenesis, with N-acetylgalactosamine expression detected after the animals had reached sexual maturity.
