Staphylococcus is a Gram-positive bacteria that causes infections in humans. This study aims to determine the distribution of Staphylococcus sp. based on clinical specimens, gender, age, and treatment room and to see the level of sensitivity to various types of antibiotics from January 2020 to March 2025 in the Clinical Microbiology Laboratory of Dr. Zainoel Abidin Regional Hospital. This study used a laboratory observation method by collecting data on Staphylococcus sp. culture results from various clinical specimens. Data were collected retrospectively and prospectively. Bacteria were isolated on Blood Agar media and identification and antibiotic sensitivity testing were carried out using the VITEK ® 2 Compact tool. The results of the study obtained 29 species of Staphylococcus from a total of 5,166 identified isolates. The most dominant species distribution was Staphylococcus aureus at 28.69%, followed by Staphylococcus haemolyticus and Staphylococcus hominis ssp hominis at 23.87% and 19.82%, respectively. The distribution of Staphylococcus sp. Based on clinical specimens, the most common type of Staphylococcus sp. was found in blood specimens, namely 52.86%, followed by pus specimens and fluid specimens, respectively 17.13% and 12.27%. The distribution of Staphylococcus sp. based on gender was most commonly found in men, namely 55.94% and the age range most commonly found was in the age of >59 years. namely 35.15%. The distribution of Staphylococcus sp. based on the highest treatment room was found in the ICU, namely 19.13%. The prevalence of Methicillin-Resistant Staphylocccus aureus (MRSA) in this study was 79.01%. Keywords: Culture, clinical specimen, species, Staphylococcus sp., antibiotic sensitivity
