Ekstrak kulit buah kakao (Theobroma cacao L.) telah diuji aktivitas antidiabetesnya terhadap tikus putih jantan (Rattus norvegicus) dengan menggunakan metode streptozotocin (STZ). Kulit buah kakao sebanya 500 gram dimaserasi menggunakan air, kemudian dipartisi dengan n-heksana dan diekstraksi dengan etil asetat sehingga diperoleh ekstrak Theobroma cacao air (TCW), Theobroma cacao heksana (TCH) Theobroma cacao etil asetat (TCEA) dan Theobroma cacao air hasil partisi (TCWP) berturut-turut sebanyak 135,98 g (27,19%); 0,124g (0,091%), 0,411 g (0,302%), dan 134,49 g (98,90%). Ekstrak TCEA, TCW, TCWP memiliki kadar fenolik berturut-turut sebesar 570,44; 20,88 dan 47,44 mg GAE/g serta flavonoid berturut-turut sebesar 4,47;0,94 dan 1,54 mg QE/g. Hasil uji antidiabetes ekstrak kulit buah kakao terhadap tikus putih jantan menunjukkan presentase penurunan kadar gula darah sebesar 73,6 %. Hasil uji antidiabetes juga menunujukkan ekstrak TCEA pada dosis 25,50, dan 100 mg/kgBB mampu menurunkan kadar gula darah pada tikus putih jantan dengan nilai signifikansi 0,000 (p

The extract of cocoa pod husk (Theobroma cacao L.) has been tested for its antidiabetic activity on male white rats (Rattus norvegicus) using the streptozotocin (STZ) method. A total of 500 grams of cocoa pod husk was macerated using water, then partitioned with n-hexane and extracted with ethyl acetate, resulting in the following extracts: Theobroma cacao water extract (TCW), Theobroma cacao hexane extract (TCH), Theobroma cacao ethyl acetate extract (TCEA), and partitioned Theobroma cacao water extract (TCWP), yielding respectively 135.98 g (27.19%), 0.124 g (0.091%), 0.411 g (0.302%), and 134.49 g (98.90%).The TCEA, TCW, and TCWP extracts had total phenolic contents of 570.44; 20.88; and 47.44 mg GAE/g, respectively, and flavonoid contents of 4.47; 0.94; and 1.54 mg QE/g, respectively. The antidiabetic activity test of the cocoa pod husk extract on male white rats showed a blood glucose reduction percentage of 73.6%.The antidiabetic test also showed that TCEA extract at doses of 25, 50, and 100 mg/kgBW was able to significantly reduce blood glucose levels in male white rats with a significance value of 0.000 (p < 0.05). The 100 mg/kgBW dose was the most effective, even showing a higher average percentage and similar significance value compared to glibenclamide, a commonly used generic antidiabetic drug.
