Osteoarthritis genu merupakan salah satu kondisi yang sering menyebabkan nyeri lutut pada pasien usia lanjut. Proloterapi dekstrosa hipertonik telah dikenal sebagai terapi yang dapat mengurangi nyeri dan meningkatkan fungsi sendi. Penelitian ini bertujuan untuk menilai efektivitas proloterapi dekstrosa hipertonik berdasarkan perubahan skor Western Ontario and McMaster Universities Arthritis Index (WOMAC) dan Numeric Rating Scale (NRS) pada pasien osteoarthritis genu di RSUDZA Banda Aceh. Penelitian ini menggunakan desain kohort retrospektif dengan pendekatan analitik observasional. Data diperoleh dari rekam medis pasien di RSUDZA Banda Aceh dan Divisi Nyeri Poliklinik Saraf. Sebanyak 56 pasien yang memenuhi kriteria inklusi dipilih menggunakan teknik total sampling. Variabel independen dalam penelitian ini adalah proloterapi dekstrosa hipertonik, sedangkan variabel dependen adalah nyeri lutut pada pasien OA Genu. Analisis data dilakukan menggunakan uji Wilcoxon signed-rank untuk membandingkan skor sebelum dan setelah tindakan proloterapi. Hasil penelitian menunjukkan bahwa mayoritas pasien OA genu adalah perempuan (69,6%), rentang indeks massa tubuh pasien OA genu terbanyak adalah 25–29,9 kg/m² (42,9%) dengan rata-rata 27.05 kg/m², kelompok usia dominan adalah 61–70 tahun (50%), dan diagnosis OA tertinggi adalah OA genu bilateral sebanyak 33 pasien (58,9%). Osteoarthritis genu is a condition that frequently causes knee pain in elderly patients. Hypertonic dextrose prolotherapy has been known as a therapy that can reduce pain and improve joint function. This study aims to assess the effectiveness of hypertonic dextrose prolotherapy based on changes in Western Ontario and McMaster Universities Arthritis Index (WOMAC) and Numeric Rating Scale (NRS) scores in genu osteoarthritis patients at RSUDZA Banda Aceh. This study used a retrospective cohort design with an observational analytical approach. Data was obtained from patient medical records at RSUDZA Banda Aceh and the Pain Division of the Neurology Polyclinic. A total of 56 patients who met the inclusion criteria were selected using total sampling techniques. The independent variable in this study was hypertonic dextrose prolotherapy, while the dependent variable was knee pain in OA Genu patients. Data analysis was carried out using the Wilcoxon signed-rank test to compare scores before and after prolotherapy. The results showed that the majority of genu OA patients were women (69.6%), the body mass index range of most genu OA patients was 25–29.9 kg/m² (42.9%) with an average of 27.05 kg/m², The dominant group age was 61–70 years (50%), and the highest diagnosis of OA was bilateral genu OA in 33 patients (58.9%).
