Testosteron merupakan hormon androgen pada hewan jantan yang berperan penting dalam sistem reproduksi terutama dalam proses spermatogenesis, sehingga peran tersebut akan berpengaruh terhadap kualitas semen. Penelitian ini bertujuan untuk mengetahui hubungan antara kadar hormon testosteron pada plasma semen dengan kualitas semen sapi aceh. Penelitian ini dilaksanakan di BPTU-HPT Indrapuri, Aceh Besar dengan menggunakan 5 ekor sapi aceh berumur 3-6 tahun dan bobot badan 300-450 kg. Semen segar dikoleksi menggunakan teknik vagina buatan serta dievaluasi secara makroskopis dan mikroskopis. Pengukuran kadar testosteron menggunakan kit ELISA testosteron komersial EIA-1887 (DRG Instruments GmbH, Jerman). Hasil penelitian menunjukkan terdapat hubungan positif antara testosteron pada plasma semen dengan gerakan massa, motilitas, viabilitas dan konsentrasi spermatozoa, sedangkan volume dan abnormalitas spermatozoa menunjukkan hubungan negatif dengan testosteron pada plasma semen. Testosterone is an androgen hormone in male animals which plays an important role in the reproductive system, especially in the spermatogenesis process, so that the role will affect the quality of semen. This study aims to determine the relationship between testosterone hormone levels in semen plasma and the quality of Aceh cattle semen. This research was carried out at BPTU-HPT Indrapuri, Aceh Besar using 5 Acehnese cattle aged 3-6 years and body weight 300-450 kg. Fresh semen was collected using an artificial vagina technique and evaluated macroscopically and microscopically. Testosterone levels were measured using the commercial testosterone ELISA kit EIA-1887 (DRG Instruments GmbH, Germany). The results showed that there was a positive relationship between testosterone in semen plasma and mass movement, motility, viability and concentration of spermatozoa, while spermatozoa volume and abnormalities showed a negative relationship with testosterone in semen plasma. Testosterone was significantly correlated with spermatozoa viability (p
