Bakteri Bacillus sp. memiliki habitat hidup yang dapat ditemukan di air, tanah, tumbuhan dan
berbagai sedimen lingkungan, yang dapat mencemari hewan dan manusia. Sebagai perantara kontak
bakteri dengan hewan salah satunya adalah melalui organ reproduksi jantan berupa preputium.
Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi bakteri Bacillus sp. pada preputium
kambing peranakan etawah dan untuk mengetahui tingkat sensitivitas antibiotik oksitetrasiklin,
amoksisilin dan vankomsin terhadap bakteri tersebut. Sampel penelitian menggunakan swab preputium
kambing peranakan etawah yang sehat secara klinis dan diperiksa menggunakan metode Carter untuk
isolasi dan identifikasi bakteri Bacillus sp. kemudian metode Kirby Bauer untuk uji sensitivitas
antibiotik oksitetrasiklin, amoksisilin dan vankomicin. Hasil pengamatan pada media Nutrient Broth
(NB) menunjukkan adanya pertumbuhan bakteri dan pada media Nutrient Agar (NA) menunjukkan
adanya pertumbuhan koloni bakteri, yang secara pewarnaan Gram berwarna ungu dan berbentuk
batang. Uji katalase dan uji glukosa serta manitol menunjukkan hasil positif, sebagai indikator bakteri
Bacillus sp. Hasil uji sensitivitas bakteri Bacillus sp. terhadap antibiotik oksitetrasiklin menunjukkan
daya hambat seluas 25,30 mm, vankomisin seluas 18,28 mm dan amoksisilin seluas 17,82 mm.
Kesimpulan penelitian ini adalah pada preputium kambing peranakan etawah ditemukan adanya bakteri
Bacillus sp. yang sensitif terhadap antibiotik oksitetrasiklin, vankomisin, dan amoksisilin.

Bacillus sp. bacteria has a living habitat that can be found in the air, soil, plants and various environmental sediments, which can contaminate animals and humans. One way of intermediary contact between bacteria and animals is through reproduction of the reproductive organ in the form of the prepuce. This research aims to isolate and identify the bacteria Bacillus sp. on the prepuce of Etawah crossbreed goats and to determine the level of sensitivity of the antibiotics oxytetracycline, amoxicillin and vancomycin to these bacteria. The research samples used preputial swabs from clinically healthy Etawah crossbreed goats and were examined using the Carter method for isolation and identification of Bacillus sp. bacteria. then the Kirby Bauer method for sensitivity testing for the antibiotics oxytetracycline, amoxicillin and vancomycin. The results of observations on Nutrient Broth (NB) media showed the growth of bacteria and on Nutrient Agar (NA) media showed the growth of bacterial colonies, which were purple in Gram stain and rod-shaped. The catalase test and glucose and mannitol tests showed positive results, as indicators of Bacillus sp. Bacillus sp. sensitivity test results. against the antibiotic oxytetracycline showed an inhibitory effect of 25.30 mm, vancomycin of 18.28 mm and amoxicillin of 17.82 mm. The conclusion of this research is that in the prepuce of Etawah crossbreed goats, Bacillus sp. bacteria were found. who are sensitive to the antibiotics oxytetracycline, vancomycin, and amoxicillin.
