Peningkatan kualitas spermatozoa setelah pemberian PGF2 secara in vivo diduga melalui peningkatan sekresi testosteron. Peningkatan testosteron setelah pemberian PGF2α kemungkinan terkait dengan stimulasi pembentukan cylic adenosine monophosphate (cAMP). Penelitian ini bertujuan mengetahui pengaruh perbedaan interval pemberian PGF2α dengan waktu pengukuran terhadap konsentrasi cAMP tikus Wistar. Dalam penelitian ini digunakan 12 tikus Wistar (Rattus norvegicus) jantan. Seluruh tikus diadaptasikan selama dua minggu. Tikus dibagi ke dalam empat kelompok perlakuan (masing-masing n=3), yakni interval injeksi 2,5 mg/kg BB PGF2α dengan koleksi darah untuk pemeriksaan cAMP selama 30 menit (P1), 60 menit (P2), 90 menit (P3), dan 120 menit (P4). Konsentrasi cAMP dalam plasma diukur menggunakan metode enzyme linked immunosorbent assay (ELISA). Data dianalisis dengan analisis varian satu arah dan dilanjutkan dengan uji Duncan. Konsentrasi cAMP pada P1; P2; P3; dan P4 masing-masing adalah 28,947±1,556; 18,34±3,279; 19,725±0,353; dan 17,667±2,773 pmol/ml. Konsentrasi cAMP pada kelompok P1 menunjukkan perbedaan yang signifikan (P0,05). Disimpulkan bahwa perbedaan interval pemberian PGF2α dengan waktu koleksi darah memiliki pengaruh terhadap konsentrasi cAMP tikus Wistar dengan konsentrasi tertinggi diperoleh pada interval 30 menit.

The increase in spermatozoa quality after administration of PGF2α in vivo is thought to be due to testosterone secretion. The increase in testosterone after PGF2α administration is likely related to the stimulation of cyclic adenosine monophosphate (cAMP) formation. This study aims to determine the effect of different PGF2α administration intervals and measurement times on increasing cAMP concentrations in Wistar rats. In this study, 12 male Wistar rats (Rattus norvegicus) were used. Rats were adapted for two weeks and then divided into four treatment groups (n=3 each), namely injection intervals of 2.5 mg/kg BW PGF2α with blood collection for cAMP examination for 30 minutes (P1), 60 minutes (P2), 90 minutes (P3), and 120 minutes (P4). The cAMP concentration in plasma was measured using the enzyme linked immunosorbent assay (ELISA) method. Data were analyzed using one-way analysis of variance and followed by Duncan's test. The concentration of cAMP at P1; P2; P3; and P4 were 28.947±1.556; 18.34 ± 3.279; 19.725 ± 0.353; and 17,667±2,773 pmol/mL, respectively. The cAMP concentration in group P1 showed a significant difference (P0.05). It was concluded that the difference between the interval of administration of PGF2α and the time of blood collection had the cAMP concentration in Wistar rats with the highest concentration obtained at an interval of 30 minutes.
