Glikokonjugat berperan penting dalam proses diferensiasi dan maturasi sel, pengenalan dan adhesi terhadap sel, serta interaksi antar sel. Keberadaan glikokonjugat di jaringan epididimis diduga berperan dalam membantu proses pematangan (maturasi) spermatozoa. Penelitian ini bertujuan mengetahui distribusi glikokonjugat dengan residu gula N-asetilgalaktosamin pada caput, corpus, dan cauda epididimis sapi aceh yang dideteksi menggunakan pewarnaan histokimia lektin peanut agglutinin (PNA). Penelitian ini menggunakan lima epididimis sinister dari lima ekor sapi aceh jantan dewasa berumur sekitar dua tahun yang dikoleksi dari Rumah Potong Hewan Lambaro, Aceh Besar. Sampel diproses menjadi preparat histologi dan diwarnai dengan pewarnaan lektin PNA metode avidin biotin complex peroxidase (ABC method). Distribusi N-asetilgalaktosamin dianalisis secara deskriptif, sedangkan perbedaan ekspresi N-asetilgalaktosamin dinilai dengan pemberian skor intensitas (intensity score). Hasil skor dianalisis menggunakan uji Kruskal-Wallis dan uji lanjut Mann Whitney U test. Hasil penelitian menunjukkan bahwa N-asetilgalaktosamin terdistribusi pada akrosom yang terdapat di proksimal kepala spermatozoa yang berada di dalam lumen caput, corpus, dan cauda epididimis sapi aceh. Glycoconjugates play an important role in the process of cell differentiation and maturation, recognition and adhesion to cells, and interactions between cells. The presence of glycoconjugates in epididymal tissue is thought to play a role in assisting the maturation process of spermatozoa. This study aimed to determine the distribution of glycoconjugates with N-acetylgalactosamine sugar residues in the caput, corpus, and cauda epididymis of Aceh cattle detected using peanut agglutinin (PNA) lectin histochemical staining. This study used five sinister epididymis from five adult male aceh cattle aged about two years collected from Lambaro Slaughterhouse, Aceh Besar. The samples were processed into histology preparations and stained with avidin biotin complex peroxidase (ABC method) PNA lectin staining. The distribution of N-acetylgalactosamine was analyzed descriptively, while differences in N-acetylgalactosamine expression were assessed by giving an intensity score. The scores were analyzed using Kruskal-Wallis test and Mann Whitney U test. The results showed that N-acetylgalactosamine was distributed in the acrosome located proximal to the spermatozoa head in the lumen of the caput, corpus, and cauda epididymis of Aceh cattle. Based on statistical tests, there was a significant difference (P < 0.05) in the expression of N-acetylgalactosamine on the head of spermatozoa in the lumen of the corpus and cauda with spermatozoa in the lumen of the caput epididymis. It is concluded that N-acetylgalactosamine can be detected in the acrosome of spermatozoa in the lumen of the caput, corpus, and cauda of the epididymis using PNA lectin with the strongest expression found in the acrosome of spermatozoa in the lumen of the corpus and cauda of the epididymis.
