Pogostemon cablin Benth merupakan salah satu tumbuhan obat yang memiliki kemampuan sebagai penyembuhan luka. Penelitian ini telah dilakukan untuk memformulasikan dan mengkarakterisasi sediaan hidrogel film-patchouli oil-perak nanopartikel (PVA/CS/PO/AgNPs) menggunakan bahan aktif minyak nilam dan perak nanopartikel hasil green synthesis menggunakan ekstrak daun nilam (Pogostemon cablin Benth). Hasil analisis GCMS minyak nilam fraksi berat mengandung patchouli alcohol sebesar 60,66% dan fraksi ringan sebesar 28,68%. Simplisia daun nilam mengandung kadar air sebesar 8,21%, kadar sari larut air sebesar 10,87%, kadar sari larut etanol sebesar 5,53% dan kadar abu total 6,35%. Analisis GCMS ekstrak air daun nilam (EADN) mengandung senyawa patchouli alcohol sebesar 71,36% sedangkan ekstrak metanol daun nilam (EMDN) mengandung senyawa patchouli alcohol sebesar 30,91%. Karakterisasi AgNPs EADN dan EMDN dianalisis menggunakan UV-VIS, FTIR, XRD dan SEM. Analisis UV-VIS AgNPs EADN dan EMDN terbentuk pada serapan panjang gelombang 400 nm. Analisis SEM menunjukkan bahwa EMDN memiliki partikel yang seragam di seluruh permukaannya, cenderung jelas dan mampu mencegah terbentuknya gumpalan dibandingkan dengan EADN. Analisis XRD didapatkan ukuran kristal perak pada AgNPs EADN sebesar 7,22 nm dan AgNPs EMDN sebesar 20,335 nm. Analisis FTIR memperlihatkan adanya ikatan hidrogen antara gugus fungsi dari PVA, CS, dan PO. Analisis SEM menunjukkan bahwa hidrogel film yang dihasikan tidak tidak mengalami keretakan (cracking) atau lobang (pinholes) dan sedikit mengalami agglomerasi. Hasil evaluasi sediaan uji organoleptis menunjukkan bahwa sediaan hidrogel berbau khas nilam, semi solid, homogen, berwarna coklat dan ungu pada masinng-masing ekstrak. Uji daya sebar menunjukkan setiap formulasi berukuran antara 1,5-2,9 cm. Uji pH menunjukkan bahwa pH setiap formulasi berkisar antara 6,03-6,67. Uji fraksi gel dan rasio sweeling setiap formulasi masing-masing berkisar antara 1,88-2,96% dan 1,07-1,50 %.

Pogostemon cablin Benth is a medicinal plant that has the ability to heal wounds. This research was conducted to formulate and characterize film-patchouli oil-silver nanoparticle hydrogel preparations (PVA/CS/PO/AgNPs) using the active ingredients patchouli oil and silver nanoparticles resulting from green synthesis using patchouli leaf extract (Pogostemon cablin Benth). The results of the GCMS analysis of patchouli oil in the heavy fraction contained 60.66% patchouli alcohol and 28.68% in the light fraction. Patchouli leaf simplicia contains a water content of 8.21%, a water-soluble extract content of 10.87%, an ethanol-soluble extract content of 5.53% and a total ash content of 6.35%. GCMS analysis of patchouli leaf aqueous extract (EADN) contains 71.36% patchouli alcohol compound while patchouli leaf methanol extract (EMDN) contains 30.91% patchouli alcohol compound. Characterization of EADN and EMDN AgNPs was analyzed using UV-VIS, FTIR, XRD and SEM. UV-VIS analysis AgNPs EADN and EMDN formed at an absorption wavelength of 400 nm. SEM analysis shows that EMDN has uniform particles throughout its surface, tends to be clear and is able to prevent the formation of agglomerates compared to EADN. XRD analysis showed that the size of silver crystals in AgNPs EADN was 7.22 nm and AgNPs EMDN was 20.335 nm. FTIR analysis showed the presence of hydrogen bonds between the functional groups of PVA, CS, and PO. SEM analysis showed that the resulting hydrogel film did not experience cracking or pinholes and slightly agglomerated. The results of the evaluation of the organoleptic test preparations showed that the hydrogel preparations had a distinctive patchouli odor, semi-solid, homogeneous, brown and purple in each extract. The spreadability test showed that each formulation was between 1.5-2.9 cm in size. The pH test showed that the pH of each formulation ranged from 6.03-6.67. Gel fraction test and sweeling ratio for each formulation ranged between 1.88-2.96% and 1.07-1.50%, respectively.
