Abstrak balakacida (chromolaena odorata) merupakan tanaman tingkat tinggi yang secara empiris digunakan sebagai obat tradisional. tanaman tingkat tinggi diduga memiliki bakteri endofit yang mampu menghasilkan senyawa metabolit sekunder yang dapat disandikan oleh gen poliketida sintase. gen poliketida sintase menyandi enzim multifungsi yang bertanggunjawab dalam proses biosintesis poliketida sebagai senyawa bioaktif antimikroba. penelitian ini bertujuan untuk mendeteksi gen poliketida sintase sebagai penanda biosintesis antibiotik pada bakteri endofit balakacida asal banda aceh. sampel penelitian yang digunakan adalah 2 isolat bakteri endofit balakacida yang memiliki daya antibakteri kuat terhadap bakteri staphylococcus aureus, bacillus subtilis, pasteurella multocida dan pseudomonas aeruginosa yaitu beb-4 dan beb-5. penelitian dilakukan dengan tahapan diantaranya isolasi dna menggunakan prestotm mini gdna bacteria kit (geneaid), penggandaan fragmen penyandi ranah ks gen poliketida sintase menggunakan polymerase chain reaction (pcr), elektroforesis hasil pcr, sekuensing dengan menggunakan jasa perusahaan first base co. malaysia dan analisis keragaman gen berdasarkan genbank database untuk dilakukan basic local alignment search tool nucleotide (blastn) pada national center of biotechnology information (ncbi) serta konstruksi pohon filogenetik menggunakan software mega x. hasil penelitian menunjukkan domain ks gen poliketida sintase berhasil diamplifikasi dari beb-4 dan beb-5. berdasarkan pita yang dilihat, sekuensing hanya dilakukan pada isolat beb-5. hasil analisis kemiripan urutan nukleotida dan filogenetik menunjukkan bahwa isolat beb-5 memiliki tingkat homologi dengan bakteri stenotrophomonas sp. lm091 sebesar 92,37%. abstract chromolaena odorata is a high plant emphirically used as traditional medicines. high plants generally have endophytic bacteria that are capable of producing secondary metabolites compounds that can be encoded by the polyketide synthase gene. the polyketide synthase gene encode a multifunctional enzyme that is responsible for the biosynthesis of polyketides in bioactive antimicrobial ability. this research aims to detect the polyketide synthase gene as a marker of antibiotic biosynthetic of endophytic bacteria isolated from balakacida from banda aceh. samples used were beb-4 and beb-5, too endophytic bacteria isolated from c. odorata with strong antibacterial activity against staphylococcus aureus, bacillus subtilis, pasteurella multocida and pseudomonas aeruginosa. the research design was carried out in several steps, including dna isolation using the prestotm mini gdna bacteria kit (geneaid), dna amplification using polymerase chain reaction (pcr), electrophoresis of pcr results, sequencing using the services from first base co. company, malaysia and gene diversity analysis based on genbank database for basic local alignment search tool nucleotide (blastn) at the national center of biotechnology information (ncbi) and phylogenetic tree construction using mega x software. the results showed that the ks domain of polyketide synthase gene was successfully amplified from beb-4 and beb-5. based on dna band, sequencing was only carried out on beb-5. the results of the analysis of the similarity of nucleotide sequences and phylogenetic showed that the beb-5 isolate shemed 92,37% homology with the genus stenotrophomonas sp. lm091.
