Abstrak metabolit sekunder daun pala diisolasi dengan maserasi 3500 g serbuk daun pala menggunakan pelarut metanol dan diperoleh ekstrak sebanyak 535,7 g (15,3%) dengan nilai ic 50 188,4 ppm, dilanjutkan partisi dengan pelarut n-heksana, etil asetat dan kloroform, diperoleh ekstrak masing-masing 1,8488 g (0,35%), 0,501g (0,15%) dan 7,122 g (2,28%) dengan nilai ic 50 96,67 ppm, 0,216 ppm dan 35,62 ppm. ekstrak nheksana dan kloroform diisolasi lebih lanjut dengan kromatografi kolom gravitasi menggunakan eluen masing-masing n-heksana : etil asetat (9:1) dan kloroform : etil asetat (8:2) dengan hasil diperoleh masing-masing 7 fraksi gabungan (mfhd 1-7) dan 6 fraksi gabungan (mfkd 1-6) dimana seluruh fraksi gabungan aktif sebagai antioksidan. fraksi gabungan dengan aktivitas tertinggi yaitu mfhd 2 (ic 50 96,716 ppm), mfhd 4 (ic 50 0,729 ppm), mfkd 4 (ic 50 69,132 ppm) dan mfkd 5 (ic 50 direkromatografi dan dimurnikan dengan klt preparatif, diperoleh fraksi dengan pola noda tunggal pada kromatogram klt yaitu mfhd 2.3 (5,4 mg), mfhd 4.4 (10,7 mg) dan mfkd 4.4 (5,8 mg) dengan ic 50 masing-masing 42,299 ppm, 44,084 ppm dan 111,91 ppm. hasil analisis gc-ms fraksi mfhd 2.3 mengandung senyawa eucalyptol dan sitostenone. berdasarkan beberapa referensi kedua senyawa tersebut memiliki aktivitas antioksidan. kata kunci: daun pala (myristica fragrans houtt), aktivitas antioksidan 80,44 ppm) abstract isolation of secondary metabolites nutmeg leaf maceration begins with 3500 g nutmeg leaf powder using methanol and obtained extract as much as 535.7 g (15.3%) with ic 50 value of 188.4 ppm, followed partition with n-hexane, ethyl acetate and chloroform, each extract was obtained 1.8488 g (0.35%), 0,501g (0.15%) and 7.122 g (2.28%) with ic 50 value of 96.67 ppm, 0.216 ppm and 35.62 ppm. the extract n-hexane and chloroform were isolated further by column chromatography gravity using eluent each n-hexane : ethyl acetate (9 : 1) and chloroform : ethyl acetate (8 : 2) with the results obtained by each of the 7 fractions combined (mfhd 1-7) and 6 fractions combined (mfkd 1-6) in which all of the combined fractions are active as antioxidants. the combined fraction with the highest activity mfhd 2 (ic50 96.716 ppm), mfhd 4 (ic50 0,729 ppm), mfkd 4 (ic50 69.132 ppm) and mfkd 5 (ic50 80.44 ppm) return column chromatography gravity and purified by preparative tlc, the fraction with a pattern single spot on tlc chromatograms are mfhd 2.3 (5.4 mg), mfhd 4.4 (10.7 mg) and mfkd 4.4 (5.8 mg) with ic 50 respectively 42.299 ppm, 44.084 ppm and 111.91 ppm. the gc-ms analysis of mfhd 2.3 fraction contains eucalyptol and sitostenone compounds. based on some references both of these compounds have antioxidant activity. keywords: nutmeg leaf (myristica fragrans houtt), antioxidant activity
