Disertasi ini ditulis oleh rika husna dengan judul “ pengembangan formulasi bioenkapsulasi rizobakteri indigenous pgpr lokal aceh sebagai inovasi biofertilizer berkelanjutan untuk peningkatan pertumbuhan dan hasil bawang merah (allium ascalonicum. l.). penelitian ini dilaksanakan dibawah bimbingan prof. dr. ir. hasanuddin, m.s. sebagai promotor, serta prof. dr. ir. rina sriwati, m.si. dan dr. sulastri, s.p., m.si. sebagai co-promotor. disertasi ini membahas isu strategis dalam pengembangan pertanian berkelanjutan, khususnya terkait dengan penurunan produksi bawang merah (allium ascalonicum. l). permintaan bawang merah terus meningkat, namun produksi di aceh mengalami penurunan, kondisi ini menandakan adanya kesenjangan antara kebutuhan dan ketersediaan bawang merah. penurunan produksi disebabkan oleh rendahnya kesuburan tanah, keterbatasan hara, serta minimnya mikroorganisme bermanfaat di daerah perakaran. tingkat kelangsungan hidup pgpr secara biologis di tanah tidak stabil dan aktivitas bakteri menurun dari waktu ke waktu karena faktor lingkungan, bioenkapsulasi merupakan solusi yang menjanjikan, karena mampu meningkatkan viabilitas, stabilitas, serta pelepasan pgpr secara terkendali. penelitian ini bertujuan untuk (1) meningkatkan stabilitas pgpr, mengontrol pelepasan pgpr yang telah dikapsulasikan, (2) mengukur pengaruh bioenkapsulasi pgpr terhadap pertumbuhan dan hasil bawang merah, dan (3) membandingkan efektivitas bioenkapsulasi pgpr dengan aplikasi pgpr konvensional. penelitian dilakukan dalam tiga tahap, tahap pertama adalah skrining isolat rizobakteri yang berpotensi sebagai plant growth promoting rhizobacteria (pgpr) pada benih tanaman bawang merah secara invitro, yang kemudian dikarakterisasi berdasarkan morfologi dan fisiologi, termasuk kemampuan menghasilkan iaa, siderofor, hcn, acc deaminase, fiksasi n serta pelarutan fosfat dan pelarutan kalium. tahap kedua adalah pengujian formula bahan penyalut yang digunakan pada proses bioenkapsulasi dalam meningkatkan stabilitas dan mengontrol pelepasan kapsulasi pgpr. tahap ketiga adalah pengaplikasian bioenkapsulasi pgpr terhadap pertumbuhan dan hasil bawang merah. tahap keempat adalah mengidentifikasi molekular isolat rizobakteri indigenous asal rizosfer bawang merah. hasil penelitian tahap pertama menunjukkan bahwa eksplorasi dan karakterisasi fisiologis dan morfologis berhasil mengidentifikasi isolat rizobakteri yang berpotensi sebagai plant growth promoting rhizobacteria (pgpr). hal ini ditunjukkan oleh kemampuan menghasilkan senyawa bioaktif dan nutrisi penting bagi tanaman. tahap kedua, formula bahan penyalut kombinasi antara alginat dan kitosan memberikan pengaruh yang sangat nyata terhadap sifat fisik dan kimia bioenkapsulasi. tahap ketiga, bioenkapsulasi isolat rizobakteri terpiih signifikan meningkatkan pertumbuhan dan hasil bawang merah. tahap keempat, keberhasilan identifikasi molekular isolat unggulan menunjukkan kekerabatan erat dengan spesies bakteri yang dikenal sebagai agens biofertilizer dan bioprotektan, seperti bacillus subtilis, pseudmonas aeruginosa, brevibacillus brevis, bacillus amyloliquefaciens. kesimpulan pertama, ditemukan 31 isolat rizobakteri indigenous dari rizosfer bawang merah aceh yang terbukti memiliki karakter morfologi dan fisiologis yang berperan sebagai pgpr. kedua, formula bahan penyalut alginat 1%:kitosan 1,5% menunjukkan hasil terbaik pada stabilitas agregat dan kadar air bioenkapsulasi. kelarutan protein tertinggi pada formula alginat 1%:kitosan 0,5%, dan efisiensi bioenkpsulasi tertinggi pada formula alginat 1%:kitosan 1%. ketiga, aplikasi bioenkapsulasi pgpr signifikan meningkatkan pertumbuhan dan hasil bawang merah. disarankan dilakukan analisis lanjutan dengan pendekatan metagenomik untuk memahami mekanisme kolonisasi, komunikasi antara tanaman-mikroba, serta respon tanaman secara menyeluruh terhadap inokulasi pgpr yang telah ditransformasikan melalui metode bioenkapsulasi.

Abstract
This dissertation authored by Rika Husna and is titled “Development of a Bioencapsulated Formulation of Indigenous Rhizobacteria from Aceh as a Sustainable Biofertilizer Innovation to Increase Growth and Yield of Shallots (Allium ascalonicum L.)” This research was conducted under the supervision of Prof. Dr. Ir. Hasanuddin, M.S., as the Promoter, and Prof. Dr. Ir. Rina Sriwati, M.Si., and Dr. Sulastri, S.P., M.Si., as Co-Promoters. This dissertation addresses strategic challenges in the development of sustainable agriculture, with a particular focus on the decline in shallot (Allium ascalonicum L.) production in Aceh. Although demand for shallots continues to rise, local production has experienced a significant decrease, creating a gap between supply and demand. This decline is largely attributed to low soil fertility, nutrient limitations, and the absence of beneficial microorganisms in the rhizosphere. Furthermore, the biological survival rate of plant growth-promoting rhizobacteria (PGPR) in soil is unstable, as bacterial activity diminishes over time due to environmental stress. Bioencapsulation has emerged as a promising solution to these challenges, offering enhanced viability, stability, and controlled release of PGPR, thereby improving their effectiveness in agricultural applications. The study was designed to achieve four main objectives: improving the stability and regulating the release of encapsulated PGPR, evaluating the effects of PGPR bioencapsulation on shallot growth and yield, comparing the effectiveness of bioencapsulated PGPR with conventional PGPR application, and identifying the molecular characteristics and phylogenetic relationships of indigenous rhizobacterial isolates from the shallot rhizosphere in Aceh. Research was conducted in sequential stages. The first stage involved the exploration and characterization of rhizobacteria from shallot rhizosphere soil, focusing on morphological and physiological traits such as IAA production, siderophore formation, HCN release, ACC deaminase activity, nitrogen fixation, and phosphate and potassium solubilization. The second stage tested coating formulations for bioencapsulation, particularly combinations of alginate and chitosan, to enhance capsule stability and regulate PGPR release. The third stage applied bioencapsulated PGPR under experimental conditions to assess its impact on shallot growth and yield. The fourth stage involved molecular identification of superior isolates using 16S rRNA sequencing and phylogenetic analysis. The findings demonstrate that thirty-one indigenous rhizobacterial isolates were successfully identified, exhibiting morphological and physiological traits consistent with PGPR functionality. These isolates produced bioactive compounds and nutrients essential for plant growth, confirming their potential as PGPR. The alginate–chitosan coating formulations significantly influenced encapsulation performance, with the 1% alginate:1.5% chitosan formulation providing the best aggregate stability and water retention, the 1% alginate:0.5% chitosan formulation yielding the highest protein solubility, and the 1% alginate:1% chitosan formulation achieving the highest encapsulation efficiency. Application of bioencapsulated PGPR markedly improved shallot growth and yield compared to conventional PGPR application, confirming the superior performance of encapsulation technology. Molecular identification revealed close phylogenetic relationships between Aceh isolates and globally recognized PGPR species such as Bacillus subtilis, Bacillus amyloliquefaciens, Pseudomonas aeruginosa, and Brevibacillus brevis, strengthening the evidence that indigenous isolates can serve as biofertilizer, biostimulant, and bioprotectant agents. In conclusion, this dissertation provides a novel contribution by developing and validating bioencapsulation technology for indigenous PGPR isolates from Aceh. The study not only demonstrates the effectiveness of encapsulated PGPR in enhancing shallot productivity but also establishes a molecular and phylogenetic basis for their application as sustainable biofertilizers. These findings highlight the potential of indigenous PGPR bioencapsulation as an innovative approach to address declining shallot production and to promote sustainable agricultural practices. Future research employing metagenomic approaches is recommended to further elucidate the mechanisms underlying PGPR–plant interactions and to optimize biofertilizer formulations for broader agricultural applications.
