Disertasi ini meneliti potensi ekstrak buah melia azedarach l. sebagai agen antidiabetes berbasis bahan alam yang dikaji secara ilmiah melalui pendekatan terpadu fitokimia, biokimia, dan farmakologi. penelitian ini berawal dari pemanfaatan tradisional buah m. azedarach l. oleh masyarakat aceh dalam pengobatan diabetes mellitus, yang hingga saat ini belum didukung oleh kajian ilmiah yang komprehensif dan sistematis, khususnya pada bagian buah tanaman, karena sebagian besar penelitian sebelumnya lebih berfokus pada daun, kulit batang, atau biji. kebaruan (novelty) penelitian ini terletak pada integrasi analisis profil senyawa bioaktif buah m. azedarach l. berdasarkan perbedaan polaritas pelarut, evaluasi aktivitas antioksidan, pengujian penghambatan dua enzim kunci metabolik (α-amilase dan hmg-coa reduktase) secara in vitro, serta pembuktian efek hipoglikemik secara in vivo dalam satu kerangka penelitian yang utuh. penelitian ini merupakan kajian pertama yang secara khusus mengaitkan komposisi metabolit sekunder buah m. azedarach l. dengan mekanisme kerja multitarget dalam pengendalian diabetes mellitus berbasis kearifan lokal aceh. tujuan penelitian ini adalah untuk mengidentifikasi senyawa bioaktif, mengevaluasi aktivitas antioksidan, menguji kemampuan penghambatan enzim metabolik, serta menganalisis efek hipoglikemik secara in vivo dari ekstrak buah m. azedarach l. ekstraksi dilakukan menggunakan tiga jenis pelarut dengan tingkat kepolaran berbeda, yaitu n-heksana, etil asetat, dan metanol dalam memperoleh profil senyawa yang representatif. metode penelitian meliputi analisis fitokimia kualitatif menggunakan kromatografi lapis tipis (klt), penetapan kandungan total fenolik dan flavonoid, serta uji aktivitas antioksidan metode dpph. karakterisasi senyawa bioaktif dilakukan menggunakan gas chromatography mass spectrometry (gcms). selanjutnya, ekstrak dengan aktivitas biologis terbaik diuji secara in vitro melalui penghambatan enzim α-amilase dan hmg-coa reduktase, serta secara in vivo pada tikus hiperglikemik untuk mengamati perubahan kadar glukosa darah. hasil penelitian menunjukkan bahwa ekstrak buah m. azedarach l. mengandung berbagai metabolit sekunder penting, terutama fenolik, flavonoid, terpenoid, dan steroid, dengan aktivitas antioksidan serta penghambatan enzim metabolik yang signifikan. pemberian ekstrak pada tikus hiperglikemik mampu menurunkan kadar glukosa darah serta memperbaiki kondisi jaringan metabolik, yang mengindikasikan mekanisme kerja multitarget dalam pengendalian diabetes mellitus. disertasi ini memberikan kontribusi ilmiah berupa penguatan bukti ilmiah terhadap pemanfaatan buah m. azedarach l. sebagai sumber kandidat antidiabetes alami, sekaligus memperluas dasar ilmiah pengembangan fitofarmaka berbasis kearifan lokal aceh. hasil penelitian ini diharapkan menjadi landasan dalam pengembangan obat herbal terstandar serta mendorong penelitian lanjutan di bidang farmasi, biomedik, dan kesehatan masyarakat. kata kunci : melia azedarach l., buah mindi, antidiabetes, metabolit sekunder, antioksidan, penghambatan enzim, gcms, diabetes mellitus.



Abstract

This dissertation investigates the potential of Melia azedarach L. fruit extract as a natural-based antidiabetic agent through an integrated phytochemical, biochemical, and pharmacological approach. The study originates from the traditional use of M. azedarach L. fruit by the Acehnese community in the treatment of diabetes mellitus, which has not yet been supported by comprehensive and systematic scientific evidence, particularly focusing on the fruit, as most previous studies have predominantly examined the leaves, bark, or seeds. The novelty of this research lies in the integration of bioactive compound profiling of M. azedarach L. fruit based on solvent polarity differences, evaluation of antioxidant activity, in vitro inhibition of two key metabolic enzymes (α-amylase and HMG-CoA reductase), and in vivo validation of hypoglycemic effects within a single comprehensive research framework. This study represents the first investigation that specifically correlates the secondary metabolite composition of M. azedarach L. fruit with a multitarget mechanism of action in the management of diabetes mellitus, grounded in the local wisdom of Aceh. The objectives of this study were to identify bioactive compounds, evaluate antioxidant activity, assess metabolic enzyme inhibitory potential, and analyze the in vivo hypoglycemic effects of M. azedarach L. fruit extracts. Extraction was performed using three solvents with different polarity levels, namely n-hexane, ethyl acetate, and methanol, to obtain representative phytochemical profiles. The research methods included qualitative phytochemical analysis using Thin Layer Chromatography (TLC), determination of total phenolic and flavonoid contents, and antioxidant activity assessment using the DPPH method. Bioactive compound characterization was conducted using Gas Chromatography Mass Spectrometry (GCMS). Furthermore, the extract exhibiting the highest biological activity was evaluated in vitro for its inhibitory effects on α-amylase and HMG-CoA reductase, and in vivo using hyperglycemic rat models to observe changes in blood glucose levels. The results demonstrated that M. azedarach L. fruit extracts contain various important secondary metabolites, particularly phenolics, flavonoids, terpenoids, and steroids, which exhibited significant antioxidant activity and metabolic enzyme inhibition. Administration of the extract to hyperglycemic rats effectively reduced blood glucose levels and improved metabolic tissue conditions, indicating a multitarget mechanism in the regulation of diabetes mellitus. This dissertation provides scientific evidence supporting the traditional use of M. azedarach L. fruit as a potential natural antidiabetic agent and expands the scientific foundation for the development of phytopharmaceuticals based on Acehnese local wisdom. The findings are expected to serve as a basis for the development of standardized herbal medicines and to stimulate further research in the fields of pharmacy, biomedicine, and public health.
