Penelitian ini bertujuan untuk menganalisis pengaruh penambahan propolis pada media pengencer ringer terhadap kualitas sperma ikan keudawah (rasbora arundinata) pasca kriopreservasi serta menentukan konsentrasi optimum yang mampu mempertahankan kualitas spermatozoa setelah proses pembekuan. penelitian menggunakan rancangan acak lengkap (ral) dengan tujuh perlakuan konsentrasi propolis, yaitu 0; 0,2; 0,4; 0,6; 0,8; 1,0; dan 1,2 mg/ml, masing-masing dengan tiga kali ulangan. propolis dikombinasikan dengan 10% dimetil sulfoksida (dmso) sebagai krioprotektan intraseluler, sedangkan media pengencer yang digunakan adalah larutan ringer. sampel sperma disimpan dalam nitrogen cair pada suhu −196°c selama 7 hari. parameter yang diamati meliputi motilitas total,(fast motility, slow motility, circle motility, local motility, immotile), viabilitas, abnormalitas, serta karakteristik kinetik sperma. data dianalisis menggunakan analisis varians (anova) dan dilanjutkan dengan uji duncan. hasil penelitian menunjukkan bahwa penambahan propolis memberikan pengaruh nyata (p



Abstract

This study aimed to evaluate the effects of propolis supplementation in a Ringer solution extender on the post-thaw sperm quality of Keudawah fish (Rasbora arundinata) and to determine the optimum concentration for maintaining sperm quality after cryopreservation. A completely randomized design (CRD) was employed, consisting of seven propolis concentrations (0, 0.2, 0.4, 0.6, 0.8, 1.0, and 1.2 mg/mL), with three replicates per treatment. Propolis was combined with 10% dimethyl sulfoxide (DMSO) as the intracellular cryoprotectant, while Ringer solution served as the extender. Sperm samples were cryopreserved in liquid nitrogen at −196°C for seven days. The evaluated parameters included total motility, fast motility, slow motility, circular motility, local motility, immotile sperm, viability, abnormality, and sperm kinetic characteristics. Data were analyzed using analysis of variance (ANOVA), followed by Duncan’s multiple range test. The results demonstrated that propolis supplementation significantly affected all evaluated sperm quality parameters (P < 0.05). The optimum treatment was achieved with a propolis concentration of 0.6 mg/mL, resulting in total motility of 80.46 ± 4.24%, fast motility of 5.93 ± 2.84%, circular motility of 1.61 ± 0.22%, viability of 75.43 ± 4.32%, the lowest abnormality rate (1.50 ± 0.70%), and the lowest percentage of immotile sperm (20.75 ± 2.73%). This treatment also produced the highest sperm kinetic values, including curvilinear velocity (VCL) of 62.34 ± 1.78 µm/s, straight-line velocity (VSL) of 47.55 ± 20.71 µm/s, average path velocity (VAP) of 45.78 ± 18.58 µm/s, and beat cross frequency (BCF) of 7.18 ± 6.57 Hz. In contrast, the control treatment without propolis supplementation exhibited the poorest post-thaw sperm quality. The observed improvement in sperm quality at the optimum concentration was likely attributable to the flavonoid and phenolic compounds present in propolis, which function as antioxidants and protect sperm cell membranes against oxidative damage during cryopreservation. Therefore, supplementation of Ringer solution extender with 0.6 mg/mL propolis is an effective strategy for maintaining the post-thaw quality of Keudawah fish sper.
