Penuaan kulit (photoaging) merupakan proses degeneratif kulit yang disebabkan oleh faktor intrinsik berupa penambahan usia dan faktor ekstrinsik berupa paparan sinar uv yang menyebabkan penurunan kelembapan, kerusakan elastisitas, dan kolagen kulit. salah satu upaya yang dapat dilakukan untuk mencegah kerusakan tersebut adalah pemanfaatan bahan alam yang mengandung senyawa antioksidan, seperti daun cempaka kuning (magnolia champaca l.) yang diketahui mengandung flavonoid, fenolik, dan tanin. penelitian ini bertujuan untuk mengetahui pengaruh pemberian krim esktrak n-heksan daun cempaka kuning terhadap kelembapan, elastisitas, dan kolagen kulit mencit (mus musculus l.) yang diinduksi dengan sinar uvb. penelitian ini merupakan penelitian eksperimental menggunakan rancangan acak lengkap (ral) dengan tujuh kelompok perlakuan yang terdiri dari kelompok kontrol normal (k0), kontrol negatif (k-), kontrol positif (k+), serta kelompok perlakuan dengan pengolesan krim ekstrak n heksan daun cempaka kuning dengan konsentrasi 0 x ic₅₀ (p1), 50 x ic₅₀ (p2), 100 x ic₅₀ (p3), dan 200 x ic₅₀ (p4). masing-masing kelompok terdiri atas tiga ekor mencit dengan usia 8 bulan. parameter kelembapan, elastisitas, dan kolagen kulit diukur menggunakan skin analyzer eh-900u. data dianalisis menggunakan uji shapiro-wilk, levene’s test, one-way anova, post hoc games-howell, dan kruskal-wallis sesuai dengan karakteristik distribusi data. hasil penelitian menunjukkan bahwa pemberian krim ekstrak n-heksan daun cempaka kuning meningkatkan nilai rata-rata kelembapan, elastisitas, dan kolagen kulit mencit dibandingkan dengan kontrol negatif. namun, analisis statistik menunjukkan bahwa peningkatan tersebut belum terdapat perbedaan yang signifikan (p>0,05) pada seluruh parameter, meskipun kelompok konsentrasi 100 x ic₅₀ memberikan respon biologis terbaik. berdasarkan hasil tersebut, dapat disimpulkan bahwa krim ekstrak n-heksan daun cempaka kuning berpotensi sebagai sediaan anti-aging melalui perbaikan kelembapan, elastisitas, dan kolagen kulit akibat paparan sinar uvb, meskipun belum signifikan secara statistik. kata kunci: anti-aging, ekstrak n-heksan, elastisitas kulit, kelembapan kulit, kolagen, magnolia champaca l.



Abstract

kin aging (photoaging) is a degenerative process of the skin caused by intrinsic factors such as increasing age and extrinsic factors such as UV radiation exposure, which leads to decreased skin moisture, impaired, elasticity, and collagen degradation. One effort to prevent this damage is the use of natural ingredients containing antioxidant compounds, such as yellow champaca leaves (Magnolia champaca L.) which are known to contain flavonoids, phenolic, and tannins. This study aimed to determine the effect of n-hexane extract cream from yellow champaca leaves on the moisture, elasticity, and collagen of the skin of mice (Mus musculus L.) induced by UVB radiation. This study was an experimental study using a Completely Randomized Design (CRD) with seven treatment groups consisting of a normal control group (K0), a negative control (K-), a positive control (K+), and treatment groups receiving topical application of n-hexane extract cream from yellow champaca leaves at concentrations of 0 x IC₅₀ (P1), 50 x IC₅₀ (P2), 100 x IC₅₀ (P3), and 200 x IC₅₀(P4). Each group consisted of three 8-month-old mice. Skin moisture, elasticity, and collagen parameters were measured using a Skin Analyzer EH-900U. Data were analyzed using the Shapiro-Wilk test, Levene’s Test, One-Way ANOVA, Post Hoc Games-Howell test, and Kruskal-Wallis test according to the characteristis of the data distribution. The results showed that the administration of n-hexane extract cream from yellow champaca leaves increased the mean values of skin moisture, elasticity, and collagen in mice compared with the negative control. However, statistical analysis indicated that these increases were not significantly different (p>0,05) for all parameters, although the 100 x IC₅₀ concentration group produced the best biological response. Based of these findings, it can be concluded that n-hexane extract cream from yellow champaca leaves has potential as an anti-aging formulation by impoving skin moisture, elasticity, and collagen following UVB exposure, although the effect were not statistically significant.
