Inflamasi kronis yang tidak terkendali berkontribusi terhadap berbagai penyakit degeneratif, sementara penggunaan obat antiinflamasi sintetis jangka panjang berisiko menimbulkan efek samping yang merugikan. daun srikaya (annona squamosa l.) berpotensi sebagai sumber antiinflamasi alami, namun penelitian komprehensif mengenai ekstrak airnya dengan variasi metode pengeringan masih terbatas. penelitian ini bertujuan mengevaluasi aktivitas antiinflamasi ekstrak air daun srikaya berdasarkan variasi metode pengeringan (daun segar, dikering anginkan, kering matahari, kering oven, dan kering mikrowave) dan waktu ekstraksi (1, 3, 5, 7, dan 9 menit) pada konsentrasi sampel 3 g/100 ml, menggunakan dua metode uji in vitro: inhibisi protease (ip) dan stabilisasi membran human red blood cell (hrbc). hasil menunjukkan rendemen ekstrak tertinggi diperoleh pada kelompok kering mikrowave pada waktu ekstraksi 7 menit (29,2%). pada metode ip dan hrbc, kelompok kering mikrowave menunjukkan aktivitas tertinggi dengan rata-rata ic50 berturut-turut 0,48 ppm dan 0,99 ppm, mendekati kontrol positif natrium diklofenak (0,49 dan 4,59 ppm), diikuti kelompok segar (1,01 dan 7,26 ppm). kelompok kering matahari menunjukkan aktivitas terlemah pada metode ip (10,90 ppm), sedangkan pada metode hrbc aktivitas terlemah teramati pada kelompok kering angin (47,08 ppm), berdekatan dengan kering matahari (45,65 ppm). metode ip menunjukkan proporsi titik data yang memenuhi kriteria validitas regresi (r² ≥ 0,8) sebesar 84% (21 dari 25 titik), sedangkan pada metode hrbc proporsi tersebut sebesar 76% (19 dari 25 titik), dengan titik tidak valid terkonsentrasi pada kelompok kering angin. disimpulkan bahwa kombinasi daun segar dan kering mikrowave dengan konsentrasi serbuk 3 g/100 ml menghasilkan aktivitas antiinflamasi optimal secara konsisten pada kedua metode uji.



Abstract

Uncontrolled chronic inflammation contributes to various degenerative diseases, while long-term use of synthetic anti-inflammatory drugs carries risks of gastrointestinal, cardiovascular, and nephrotoxic side effects. Sugar apple (Annona squamosa L.) leaves are a promising natural anti-inflammatory source, yet comprehensive studies on their aqueous extracts across different drying methods remain limited. This study evaluated the anti-inflammatory activity of aqueous A. squamosa leaf extract across drying methods (fresh, air-dried, sun-dried, oven-dried, microwave-dried) and extraction time (1, 3, 5, 7, 9 minutes) at a powder concentration of 3 g/100 mL, using two in vitro assays: protease inhibition (PI) and human red blood cell (HRBC) membrane stabilization. The highest extract yield was obtained from the microwavedried group at 7-minute extraction (29.2%). In the PI and HRBC assays, the microwave-dried group showed the highest activity (IC50 0.48 and 0.99 ppm, respectively), approaching the sodium diclofenac positive control (0.49 and 4.59 ppm), followed by the fresh-leaf group (1.01 and 7.26 ppm). The sun-dried group showed the weakest activity in the PI assay (10.90 ppm), while in the HRBC assay the weakest activity was observed in the air-dried group (47.08 ppm), closely followed by the sundried group (45.65 ppm). The PI method showed 84% of data points (21 of 25) meeting the regression validity criterion (R² ≥ 0.8), while the HRBC method showed 76% (19 of 25 points), with invalid points concentrated in the air-dried group. The combination of fresh or microwave-dried leaves with 3 g/100 mL powder concentration consistently produced optimal anti-inflammatory activity across both assays.
