Penelitian ini bertujuan untuk mengetahui pengaruh penambahan propolis pada media pengencer espig-01 terhadap kualitas sperma ikan keudawah (rasbora arundinata) pasca kriopreservasi. penelitian menggunakan rancangan acak lengkap (ral) dengan tujuh perlakuan konsentrasi propolis (0; 0,2; 0,4; 0,6; 0,8; 1,0; dan 1,2 mg/ml) dan tiga ulangan. propolis dikombinasikan dengan 10% dimetil sulfoksida (dmso) sebagai krioprotektan intraseluler. sampel sperma diencerkan menggunakan extender espig-01 dan disimpan dalam nitrogen cair pada suhu -196°c selama 7 hari. parameter yang diamati meliputi motilitas, fast motility, slow motility, circle motility, local motility, immotil, viabilitas, abnormalitas, dan karakteristik kinetik sperma. data dianalisis menggunakan analisis varians (anova) yang dilanjutkan dengan uji duncan. hasil penelitian menunjukkan bahwa penambahan propolis berpengaruh nyata terhadap seluruh parameter yang diamati (p

Abstract
This study aimed to determine the effect of propolis supplementation in ESPIG-01 extender on the post-cryopreservation sperm quality of keudawah fish (Rasbora arundinata). The study employed a completely randomized design (CRD) with seven propolis concentrations (0, 0.2, 0.4, 0.6, 0.8, 1.0, and 1.2 mg/mL) and three replications. Propolis was combined with 10% dimethyl sulfoxide (DMSO) as an intracellular cryoprotectant. Sperm samples were diluted with ESPIG-01 extender and stored in liquid nitrogen at −196°C for 7 days. The observed parameters included total motility, fast motility, slow motility, circular motility, local motility, immotile sperm, viability, abnormality, and sperm kinematic characteristics. Data were analyzed using analysis of variance (ANOVA) followed by Duncan’s multiple range test. The results showed that propolis supplementation significantly affected all observed parameters (P < 0.05). The best treatment was obtained at a propolis concentration of 0.6 mg/mL, which resulted in total motility of 88.75 ± 2.73%, fast motility of 5.36 ± 0.23%, circular motility of 1.45 ± 0.36%, viability of 82.25 ± 0.35%, the lowest abnormality of 1.50 ± 0.70%, and the lowest immotile sperm percentage of 11.24 ± 2.73%. This treatment also produced the highest sperm kinematic values, including curvilinear velocity (83.93 ± 7.93 µm/s), straight-line velocity (58.83 ± 7.53 µm/s), average path velocity (64.45 ± 8.57 µm/s), and beat cross frequency (12.33 ± 0.23 Hz). In contrast, the control treatment without propolis showed the lowest sperm quality. The optimum propolis concentration is presumed to protect spermatozoa against oxidative damage during freezing and thawing due to its flavonoid and phenolic compounds with antioxidant properties. In conclusion, supplementation of 0.6 mg/mL propolis in ESPIG-01 extender is effective in maintaining the post-cryopreservation sperm quality of keudawah fish.
