Abstract
Antibiotic resistance among pathogenic bacteria has become a major challenge in both veterinary and public health. This study aimed to evaluate the antibacterial activity of the halotolerant fungus Aspergillus westerdijkiae B.K 9.2 extract against Staphylococcus aureus and to determine its minimum inhibitory concentration (MIC). Antibacterial activity was evaluated using the Kirby–Bauer disk diffusion method, while the MIC was determined using the resazurin-based broth microdilution method. Preliminary screening using the disk diffusion assay without concentration standardization demonstrated antibacterial activity against Staphylococcus aureus. Subsequent testing using an extract concentration of 5000 ppm produced an average inhibition zone of 7.5 ± 0.71 mm. The MIC of the extract against Staphylococcus aureus was determined to be 5000 ppm, whereas the positive control, ampicillin, exhibited an MIC of 625 ppm. These findings indicate that the extract of the halotolerant fungus Aspergillus westerdijkiae B.K 9.2 possesses antibacterial activity against Staphylococcus aureus. Nevertheless, the extract demonstrates potential as a natural source of antibacterial compounds for further research.
