Limbah industri tahu menyimpan potensi sebagai sumber bakteri asam laktat (bal) lokal yang dapat dikembangkan menjadi probiotik, sehingga evaluasi keamanannya perlu diketahui sebelum suatu isolat ditetapkan sebagai kandidat probiotik. penelitian ini bertujuan untuk menganalisis profil resistensi antibiotik dan aktivitas hemolitik isolat bal yang berasal dari limbah padat dan cair industri tahu sebagai parameter keamanan kandidat probiotik. penelitian dilakukan terhadap tujuh isolat bal, yaitu lpt1_streptococcacea, lpt2_lactococcus lactis, lpt3_streptococcaceae, lct1_lactobacillaceae, lct2_lactobacillaceae, lct3_lactobacillaceae, dan lct5_lactococcus lactis. uji resistensi antibiotik dilakukan menggunakan metode kirby–bauer disk diffusion terhadap antibiotik tetrasiklin, gentamisin, ampisilin, dan azitromisin, sedangkan uji hemolitik dilakukan pada media blood agar. hasil penelitian menunjukkan bahwa profil resistensi antibiotik isolat bal bervariasi. isolat lct1_lactobacillaceae, lct2_lactobacillaceae, dan lct5_lactococcus lactis tidak menunjukkan kategori resisten maupun indikasi heteroresistensi, isolat lpt2_lactococcus lactis dan lpt3_streptococcaceae menunjukkan indikasi heteroresistensi, serta isolat lpt1_streptococcacea dan lct3_lactobacillaceae diasumsikan memiliki resistensi acquired. hasil uji hemolitik menunjukkan bahwa isolat lpt1_streptococcacea, lpt2_lactococcus lactis dan lpt3_streptococcaceae termasuk kategori α hemolisis, sedangkan isolat lct1_lactobacillaceae, lct2_lactobacillaceae, lct3_lactobacillaceae, dan lct5_lactococcus lactis. menunjukkan kategori γ hemolisis. berdasarkan kedua parameter keamanan tersebut, isolat lct1_lactobacillaceae, lct2_lactobacillaceae, dan lct5_lactococcus lactis memenuhi kriteria keamanan sebagai kandidat probiotik dan berpotensi untuk diteliti lebih lanjut. hasil penelitian ini mendukung pemanfaatan limbah industri tahu sebagai sumber probiotik lokal yang aman dan bernilai guna tinggi. kata kunci: aktivitas hemolitik, bakteri asam laktat, kandidat probiotik, keamanan probiotik, limbah tahu, resistensi antibiotik



Abstract

Tofu industrial waste has potential as a source of indigenous lactic acid bacteria (LAB) that can be developed as probiotics; therefore, their safety must be evaluated before being designated as probiotic candidates. This study aimed to analyze the antibiotic resistance profiles and hemolytic activities of LAB isolates derived from solid and liquid tofu industrial waste as safety parameters for probiotic candidates. Seven LAB isolates were examined: LPT1_Streptococcacea, LPT2_Lactococcus lactis, LPT3_Streptococcaceae, LCT1_Lactobacillaceae, LCT2_Lactobacillaceae, LCT3_Lactobacillaceae, and LCT5_Lactococcus lactis. Antibiotic susceptibility was assessed using the Kirby–Bauer disk diffusion method against tetracycline, gentamicin, ampicillin, and azithromycin, while hemolytic activity was evaluated on Blood Agar. The results showed that the antibiotic resistance profiles of the isolates varied. LCT1_Lactobacillaceae, LCT2_Lactobacillaceae, LCT5_Lactococcus lactis showed neither resistance nor heteroresistance, whereas LPT2_Lactococcus lactis and LPT3_Streptococcaceae exhibited heteroresistance, and LPT1_Streptococcacea and LCT3_Lactobacillaceae were presumed to possess acquired resistance. Hemolytic testing revealed that: LPT1_Streptococcacea, LPT2_Lactococcus lactis, LPT3_Streptococcaceae exhibited α-hemolysis, whereas LCT1_Lactobacillaceae, LCT2_Lactobacillaceae, LCT3_Lactobacillaceae, and LCT5_Lactococcus lactis exhibited γ-hemolysis. Based on these safety parameters, LCT1_Lactobacillaceae, LCT2_Lactobacillaceae, and LCT5_Lactococcus lactis fulfilled the safety criteria as probiotic candidates and warrant further investigation. These findings support the utilization of tofu industrial waste as a source of safe and valuable indigenous probiotic candidates.
