Heat shock protein-70 (hsp70) adalah bagian dari heat shock protein (hsp) yang bertindak dalam mekanisme penghambatan apoptosis pada sel kanker sehingga, sehingga dapat digunakan sebagai biomarker untuk proses karsinogenesis, prognosis, and respon kanker terhadap suatu terapi. penelitian ini bertujuan untuk mengevaluasi efek pemberian ekstrak etanol daun kari (murraya koenigii) selama 30 hari terhadap ekspresi hsp70 pada kanker kulit tikus putih (sprague dawley). penelitian ini menggunakan desain observasional dengan empat kelompok, yaitu kontrol negatif (p1), kontrol positif (p2), dosis 200 mg/kg bb (p3), and dosis 300 mg/kg bb (p4). ekstrak diberikan secara peroral selama 30 hari and dikoleksi menjadi sediaan blok parafin, lalu dipotong menjadi preparat slide untuk selanjutnya diwarnai menggunakan teknik imunohistokimia dengan antibodi anti-hsp70, and diamati secara histopatologi berdasarkan parameter jumlah sel yang terwarnai and intensitas pewarnaan. ekspresi hsp70 dianalisis secara semi-kuantitatif menggunakan sistem immunoreactive score (irs) and kuantitatif statistik dengan perangkat lunak spss. hasil penelitian menunjukkan bahwa pemberian terapi ekstrak etanol daun kari dapat menurunkan ekspresi hsp70 yang dibuktikan dengan uji statistik duncan. hsp70 terekspresi rendah yang normal pada kelompok p1 dengan rerata 25,44 and mengalami kenaikan sebanyak 305,03% menjadi 103,04 pada kelompok p2. penurunan yang signifikan terjadi pada kelompok p3 sebanyak 45,57% dengan rerata 56,08 and p4 turun sebanyak 51,51% dibandingkan kontrol positif, dengan rerata 49,96 yang hampir mendekati kondisi kelompok kontrol negatif p1. kata kunci: daun kari, terapi ekstrak etanol, imunohistokimia, ekspresi hsp70, dosis efektif.

Abstract
Heat Shock Protein-70 (HSP70) is a member of the Heat Shock Protein (HSP) family that acts as an anti-apoptotic shield in cancer cells; therefore, it can be used as a biomarker for carcinogenesis, prognosis, and cancer response to therapy. This study aims to evaluate the effects of administering an ethanol extract of curry leaf (Murraya koenigii) for 30 days on HSP70 expression in skin cancer in white mice (Sprague Dawley). This study employed an observational design with four groups: negative control (P1), positive control (P2), 200 mg/kg body weight (P3), and 300 mg/kg body weight (P4). The extract was administered orally for 30 days and collected as paraffin blocks, which were then sectioned into slides. These were stained using immunohistochemical techniques with anti-HSP70 antibodies and examined histopathologically based on the parameters of the number of stained cells and staining intensity. HSP70 expression was analyzed semi-quantitatively using the ImmunoReactive Score (IRS) system and statistically quantified using SPSS software. The results showed that administration of curry leaf ethanol extract therapy reduced HSP70 expression, as demonstrated by Duncan’s statistical test. HSP70 expression was low and within the normal range in group P1, with a mean of 25.44, and increased by 305.03% to 103.04 in group P2. A significant decrease occurred in group P3 by 45.57%, with a mean of 56.08, and in group P4 by 51.51% compared to the positive control, with a mean of 49.96—which was nearly identical to the levels observed in the negative control group P1.
