Kanker payudara merupakan salah satu jenis kanker dengan angka kejadian dan tingkat kematian tertinggi di dunia. pengobatan konvensional seperti kemoterapi yang saat ini banyak digunakan kerap menimbulkan resistensi, sehingga diperlukan alternatif terapi yang lebih efektif. bahan alam menjadi salah satu kandidat yang menjanjikan karena bersifat multitarget, memiliki efek samping yang lebih sedikit, serta menunjukkan efektivitas yang baik. salah satu sumber bahan alam yang belum banyak dieksplorasi adalah bakteri endofit, salah satunya bacillus sp. dari daun singkong (manihot esculenta). penelitian ini bertujuan mengidentifikasi profil senyawa aktif ekstrak metanol bakteri endofit bacillus sp. dari daun singkong berdasarkan data sekunder hasil gc-ms, serta menganalisis potensinya sebagai agen antikanker payudara melalui pendekatan in silico yang meliputi analisis network pharmacology dan molecular docking. hasil gc-ms, sebanyak 33 senyawa memenuhi kriteria untuk dikaji lebih lanjut secara in silico. analisis network pharmacology menunjukkan bahwa senyawa-senyawa aktif tersebut berpotensi sebagai agen antikanker payudara melalui penghambatan protein esr1, yang merupakan protein kunci dalam jalur pensinyalan estrogen dan menjadi target terapi utama pada kanker payudara subtipe hormone receptor-positive. analisis molecular docking dilakukan untuk mengevaluasi afinitas pengikatan masing-masing senyawa terhadap esr1, dan hasilnya menunjukkan bahwa senyawa pyrrolo[1,2-a]pyrazine-1,4-dione, hexahydro-3-(phenylmethyl)- memiliki afinitas pengikatan terbaik di antara seluruh senyawa uji, yaitu sebesar −8,0 kkal/mol. senyawa tersebut memiliki pola interaksi yang paling mirip dengan tamoxifen dibandingkan senyawa uji lainnya. kata kunci: bacillus sp. daun manihot utilissima, kanker payudara, network pharmacology, molecular docking, dan esr1



Abstract

Breast cancer is one of the most common cancers with the highest incidence and mortality rates worldwide. Conventional treatments such as chemotherapy frequently lead to drug resistance, highlighting the need for more effective therapeutic alternatives. Natural products have emerged as promising candidates due to their multitarget nature, fewer side effects, and good efficacy. One underexplored source of natural products is endophytic bacteria, microorganisms that live within plant tissues without causing disease, including Bacillus sp. isolated from cassava leaves (Manihot esculenta). This study aimed to identify the active compound profile of methanol extract from endophytic Bacillus sp. derived from cassava leaves based on secondary data from GC-MS analysis, and to evaluate its potential as a breast cancer agent through an in silico approach comprising network pharmacology and molecular docking analyses. From the GC-MS results, 33 compounds met the criteria for further in silico analysis. Network pharmacology analysis revealed that these active compounds have potential as breast cancer agents by targeting ESR1, a key protein in the estrogen signaling pathway and a primary therapeutic target in hormone receptor-positive breast cancer. Molecular docking analysis was subsequently performed to evaluate the binding affinity of each compound toward ESR1. The results demonstrated that Pyrrolo[1,2-a]pyrazine-1,4-dione, hexahydro-3-(phenylmethyl)- exhibited the highest binding affinity among all tested compounds, with a value of −8.0 kcal/mol, and showed the most similar interaction pattern to tamoxifen compared to the other compounds tested.
