Penyakit demam berdarah dengue (dbd) yang ditularkan melalui gigitan nyamuk a. aegypti masih menjadi masalah kesehatan masyarakat yang serius di indonesia. penggunaan insektisida kimia sintetis secara berulang telah memicu resistensi pada nyamuk target dan menimbulkan dampak negatif terhadap kesehatan manusia serta lingkungan. penelitian ini bertujuan untuk menganalisis efektivitas supernatan aktinobakteri tanah isolat ats11 asal stasiun penelitian soraya, kawasan ekosistem leuser, sebagai larvasida alami terhadap larva a. aegypti, serta mendeskripsikan profil metabolit sekunder dari perlakuan terbaik menggunakan analisis gc-ms. penelitian menggunakan rancangan acak lengkap (ral) faktorial dengan dua faktor perlakuan, yaitu jenis media fermentasi yeast malt broth/ymb dan yeast starch broth/ysb dan lama inkubasi (5, 10, dan 15 hari). hasil uji hayati menunjukkan bahwa perlakuan ymb secara signifikan menghasilkan mortalitas larva lebih tinggi dibandingkan ysb, dengan perlakuan ats11-ymb-10 memberikan rata-rata mortalitas tertinggi sebesar 19,5 ekor per ulangan. faktor lama inkubasi dan interaksi antara kedua faktor tidak menunjukkan pengaruh signifikan terhadap semua parameter. analisis gc-ms pada supernatan perlakuan terbaik mengidentifikasi beberapa senyawa bioaktif dengan aktivitas larvasida, di antaranya n-hexadecanoic acid, cis-vaccenic acid, palmitic acid, methyl palmitate, methyl stearate, nerol, 1-dodecanol, geranyl acetate, eugenyl acetate, .beta.-sitosterol, dan ergost-5-en-3-ol. hasil penelitian ini menunjukkan potensi supernatan ats11 sebagai alternatif larvasida biologis yang ramah lingkungan dalam pengendalian a. aegypti.



Abstract

Dengue Hemorrhagic Fever (DHF), transmitted through the bite of A. aegypti mosquitoes, remains a serious public health problem in Indonesia. Repeated use of synthetic chemical insecticides has led to resistance in target mosquito populations and caused harmful effects on human health and the environment. This study aimed to analyze the larvicidal effectiveness of the culture supernatant of soil actinobacterium isolate ATS11 from Soraya Research Station, Leuser Ecosystem Area, against A. aegypti larvae, and to characterize the secondary metabolite profile of the best treatment using GC-MS analysis. A factorial completely randomized design (CRD) was employed with two treatment factors: fermentation medium type (Yeast Malt Broth/YMB and Yeast Starch Broth/YSB) and incubation duration (5, 10, and 15 days). Bioassay results demonstrated that YMB treatments produced significantly higher larval mortality than YSB, with ATS11-YMB-10 yielding the highest mean mortality of 19.5 individuals per replicate. Incubation duration and the interaction between both factors showed no significant effect on any parameter. GC-MS analysis of the best-performing supernatant identified several bioactive compounds with larvicidal activity, including n-hexadecanoic acid, cis-vaccenic acid, palmitic acid, methyl palmitate, methyl stearate, nerol, 1-Dodecanol, geranyl acetate, eugenyl acetate, .beta.-sitosterol, and ergost-5-en-3-ol. These findings indicate the potential of ATS11 supernatant as an environmentally friendly biological larvicide for A. aegypti control.
