Acetobacter aceti merupakan bakteri yang berpotensi sebagai kandidat kultur probiotik sehingga perlu dilakukan evaluasi aspek keamanan, salah satunya melalui uji sensitivitas terhadap antibiotik. penelitian ini bertujuan untuk mengetahui sensitivitas acetobacter aceti isolat cuka aren terhadap antibiotik golongan aminoglikosida serta menilai aspek keamanan bakteri tersebut sebagai kandidat kultur probiotik. pengujian dilakukan menggunakan metode difusi cakram (kirby-bauer) dengan antibiotik kanamisin (30 µg), gentamisin (10 µg) dan streptomisin (10 µg). parameter yang diamati berupa diameter zona hambat dan interpretasi sensitivitas berdasarkan standar clinical and laboratory standards institute (clsi). hasil penelitian menunjukkan bahwa kanamisin dan gentamisin menunjukkan kategori sensitif pada seluruh pengulangan, sedangkan streptomisin menunjukkan kategori sensitif pada satu pengulangan dan kategori intermediet pada dua pengulangan. tidak ditemukan kategori resisten pada seluruh antibiotik golongan aminoglikosida yang diuji. berdasarkan hasil tersebut, acetobacter aceti isolat cuka aren masih memiliki kepekaan terhadap antibiotik golongan aminoglikosida dan mendukung keamanan bakteri tersebut sebagai kandidat kultur probiotik, khususnya dari aspek keamanan terkait resistensi antibiotik. kata kunci: acetobacter aceti, aminoglikosida, cuka aren, kandidat kultur probiotik, sensitivitas antibiotik



Abstract

Acetobacter aceti is a bacterium that has the potential to be a candidate for probiotic culture, so it is necessary to evaluate safety aspects, one of which is through a sensitivity test to antibiotics. This study aims to determine the sensitivity of Acetobacter aceti palm vinegar isolate to antibiotics of the aminoglycoside group and assess the safety aspect of these bacteria as probiotic culture candidates. The test was performed using the disc diffusion method (Kirby-Bauer) with the antibiotic’s kanamycin (30 μg), gentamycin (10 μg) and streptomycin (10 μg). The parameters observed were in the form of inhibition zone diameter and sensitivity interpretation based on Clinical and Laboratory Standards Institute (CLSI) standards. The results showed that kanamycin and gentamycin showed sensitive categories on all repetitions, while streptomycin showed sensitive categories on one repetition and intermediate categories on two repetitions. No resistance category was found in all antibiotics of the aminoglycoside group tested. Based on these results, Acetobacter aceti palm vinegar isolate still has sensitivity to aminoglycoside antibiotics and supports the safety of these bacteria as probiotic culture candidates, especially from safety aspects related to antibiotic resistance.
