Minyak nilam (pogostemon cablin) merupakan tanaman atsiri yang mengandung senyawa bioaktif yang berpotensi sebagai antioksidan dan antiinflamasi. stres oksidatif dan proses inflamasi berperan penting dalam berbagai gangguan kesehatan, sehingga diperlukan sumber bahan alam yang dapat dimanfaatkan sebagai alternatif agen terapeutik. penelitian ini bertujuan untuk mengetahui aktivitas antioksidan dan antiinflamasi ekstrak etanol 70% minyak nilam fraksi ringan dan fraksi berat. aktivitas antioksidan diuji menggunakan metode dpph dan abts, sedangkan aktivitas antiinflamasi diuji menggunakan metode penghambatan denaturasi protein. hasil uji dpph menunjukkan potensi antioksidan dengan nilai ic50 fraksi ringan (116,30 ppm), fraksi ringan fase minyak (99,29 ppm), fraksi ringan fase etanol + air (90,70 ppm), fraksi berat (88,97 ppm), fraksi berat fase minyak (73,27 ppm), dan fraksi berat fase etanol + air (14,51 ppm). hasil uji abts menunjukkan potensi antioksidan dengan nilai ic50 fraksi ringan (70,33 ppm), fraksi ringan fase minyak (73,21 ppm), fraksi ringan fase etanol + air (59,78 ppm), fraksi berat (43,87 ppm), fraksi berat fase minyak (52,39 ppm), dan fraksi berat fase etanol + air (42,03 ppm). hasil uji denaturasi protein menunjukkan nilai ic50 fraksi ringan (239,10 ppm), fraksi ringan fase minyak (176,11 ppm), fraksi ringan fase etanol + air (156,84 ppm), fraksi berat (307,16 ppm), fraksi berat fase minyak (211,27 ppm), dan fraksi berat fase etanol + air (131,14 ppm). secara keseluruhan, fraksi berat fase etanol + air menunjukkan aktivitas paling tinggi dibandingkan sampel lainnya, sehingga berpotensi dikembangkan sebagai sumber bahan alam dengan aktivitas antioksidan dan antiinflamasi. kata kunci: minyak nilam, patchouli alcohol, fraksi ringan, fraksi berat, antioksidan, antiinflamasi.



Abstract

Patchouli oil (Pogostemon cablin) is an aromatic plant that contains bioactive compounds with potential antioxidant and anti-inflammatory properties. Oxidative stress and inflammatory processes play an important role in various health disorders, so natural sources that can be used as alternative therapeutic agents are needed. This study aims to determine the antioxidant and anti-inflammatory activities of 70% ethanol extracts of light and heavy fractions of patchouli oil. Antioxidant activity was tested using the DPPH and ABTS methods, while anti-inflammatory activity was tested using the protein denaturation inhibition method. The DPPH test results showed antioxidant potential with IC50 values of light fraction (116.30 ppm), light fraction oil phase (99.29 ppm), light fraction ethanol + water phase (90.70 ppm), heavy fraction (88.97 ppm), heavy fraction oil phase (73.27 ppm), and heavy fraction ethanol + water phase (14.51 ppm). The ABTS test results showed antioxidant potential with IC50 values for the light fraction (70.33 ppm), light fraction oil phase (73.21 ppm), light fraction ethanol + water phase (59.78 ppm), heavy fraction (43.87 ppm), heavy oil phase fraction (52.39 ppm), and heavy ethanol + water phase fraction (42.03 ppm). The protein denaturation test results showed IC50 values of 239.10 ppm for the light fraction, 176.11 ppm for the light fraction of the oil phase, 156.84 ppm for the light fraction of the ethanol + water phase, 307.16 ppm for the heavy fraction, heavy oil phase fraction (211.27 ppm), and heavy ethanol + water phase fraction (131.14 ppm). Overall, the heavy ethanol + water phase fraction showed the highest activity compared to other samples, thus having the potential to be developed as a natural source with antioxidant and anti-inflammatory activity.
