Severe early childhood caries (s-ecc) merupakan karies gigi agresif pada anak usia dini dengan streptococcus mutans sebagai bakteri utama penyebabnya. pencegahan karies umumnya dilakukan melalui penggunaan pasta gigi, namun pemakaian bahan kimia dalam jangka panjang berpotensi menimbulkan efek samping sehingga diperlukan alternatif berbahan herbal. daun jamblang (syzygium cumini) diketahui mengandung senyawa bioaktif dengan aktivitas antibakteri. penelitian ini bertujuan untuk menganalisis pengaruh masa inkubasi pasta gigi ekstrak daun jamblang terhadap ph dan pertumbuhan streptococcus mutans dari isolat klinis anak s-ecc secara in vitro. penelitian ini merupakan penelitian eksperimental laboratoris dengan desain post-test only control group. isolat klinis streptococcus mutans diperoleh dari plak gigi anak s-ecc. pengukuran ph dilakukan menggunakan alat ph meter dengan mengukur ph pasta gigi ekstrak daun jamblang dengan konsentrasi 15%, 20%, dan 22,5% yang diinteraksikan streptococcus mutans pada masa inkubasi 24, 48, dan 72 jam. pertumbuhan streptococcus mutans dianalisis menggunakan metode spektrofotometri dengan mengukur optical density (od) pasta gigi hasil pengenceran dengan konsentrasi 0,9%, 1,2%, dan 1,35% pada masa inkubasi 24, 48, dan 72 jam. hasil penelitian menunjukkan bahwa ph seluruh pasta gigi stabil, sedangkan pertumbuhan streptococcus mutans menurun seiring meningkatnya konsentrasi hasil pengenceran dan masa inkubasi. kesimpulan penelitian ini adalah masa inkubasi dan konsentrasi pasta gigi ekstrak daun jamblang berpengaruh terhadap ph dan pertumbuhan streptococcus mutans pada isolat klinis anak s-ecc secara in vitro.



Abstract

Severe Early Childhood Caries (S-ECC) is an aggressive form of dental caries in young children primarily caused by Streptococcus mutans. While caries prevention is generally achieved through the use of toothpaste, the long-term use of chemical ingredients has the potential to cause side effects, necessitating herbal alternatives. Jamblang leaves (Syzygium cumini) are known to contain bioactive compounds with antibacterial activity. This study aims to analyze the effect of the incubation period of Jamblang leaf extract toothpaste on the pH and growth of Streptococcus mutans from clinical isolates of children with S- ECC in vitro. This research is a laboratory experimental study with a post-test only control group design, using clinical isolates of Streptococcus mutans obtained from the dental plaque of children with S-ECC. The pH measurements were conducted using a pH meter by measuring the pH of Jamblang leaf extract toothpaste at concentrations of 15%, 20%, and 22.5% interacted with Streptococcus mutans at incubation periods of 24, 48, and 72 hours. The growth of Streptococcus mutans was analyzed using the spectrophotometry method by measuring the Optical Density (OD) of the diluted toothpaste at concentrations of 0.9%, 1.2%, and 1.35% at the same incubation intervals. The results showed that the pH of all toothpaste samples remained stable, while the growth of Streptococcus mutans decreased as the dilution concentration and incubation period increased. The conclusion of this study is that the incubation period and concentration of Jamblang leaf extract toothpaste influence the pH and growth of Streptococcus mutans in clinical isolates of children with S-ECC in vitro.
