Deteksi kebuntingan sapi aceh umumnya masih menggunakan metode invasif seperti pengambilan darah dan palpasi rektal yang berpotensi menimbulkan stres, sehingga diperlukan metode non-invasif berbasis pengukuran metabolit progesteron feses. penelitian ini bertujuan memvalidasi dan mengaplikasikan kit elisa progesteron komersial eia-1561 (drg instruments gmbh, germany) untuk mengukur metabolit progesteron feses sapi aceh bunting secara non-invasif, mengevaluasi perbedaan konsentrasi berdasarkan umur kebuntingan, serta menganalisis korelasi dengan progesteron darah. validasi meliputi validasi analitik (uji parallelism, akurasi, dan presisi) serta validasi biologis. data parallelism dianalisis menggunakan uji korelasi, akurasi dihitung sebagai persen recovery (%recovery), presisi dinilai berdasarkan koefisien variasi (%cv) intra dan inter-assay, sedangkan perbedaan konsentrasi progesteron antar umur kebuntingan dianalisis menggunakan one way anova. hasil uji parallelism menunjukkan kurva sejajar antara standar dan sampel darah (r=0,969; p=0,031) serta sampel feses (r=0,999; p=0,001). uji akurasi menghasilkan %recovery sebesar 99%. nilai presisi menunjukkan %cv intra-assay masing-masing 2,87% untuk low quality control dan 5,64% untuk high quality control, serta %cv inter-assay 1,91% untuk low quality control dan 4,33% untuk high quality control. konsentrasi metabolit progesteron feses pada sapi aceh dengan kategori umur kebuntingan berbeda menunjukkan perbedaan sangat signifikan (p

Abstract
Pregnancy detection in aceh cattle is generally still performed using invasive methods such as blood sampling and rectal palpation, which may induce stress. Therefore, a non-invasive method based on the measurement of fecal progesterone metabolites is needed. This study aimed to validate and apply a commercial progesterone ELISA kit, EIA-1561 (DRG Instruments GmbH, Germany), to measure fecal progesterone metabolites in pregnant Aceh cattle non-invasively, to evaluate differences in metabolite concentrations based on gestational age, and to analyze the correlation with blood progesterone levels. Validation included analytical validation (parallelism, accuracy, and precision tests) and biological validation. Parallelism data were analyzed using correlation tests, accuracy was calculated as percent recovery (%recovery), and precision was assessed using intra- and inter-assay coefficients of variation (%CV). Differences in progesterone concentrations among gestational age groups were analyzed using the One Way Anova. The parallelism test showed parallel curves between the standard and blood samples (r=0.969; p=0.031) as well as fecal samples r=0.999; p = 0.001). The accuracy test produced a % recovery of 99%. Precision values showed intra-assay %CVs of 2.87% for low quality control and 5.64% for high quality control, while inter-assay %CVs were 1.91% for low quality control and 4.33% for high quality control. Fecal progesterone metabolite concentrations in aceh cattle with different gestational age categories showed highly significant differences (P < 0.05), and a positive correlation was observed between fecal and blood progesterone levels (r=0.770, p < 0.05). In conclusion, the progesterone ELISA kit EIA-1561 was analytically and biologically validated and can be used to measure fecal progesterone metabolites non-invasively, demonstrating concentration differences according to gestational age and a positive correlation between fecal and blood progesterone levels.
