Penelitian ini bertujuan untuk mengetahui efektivitas krim ekstrak etanol kulit buah kopi arabika (coffea arabica l.) dalam mempercepat penyembuhan luka bakar pada lapisan dermis kulit mencit dan menentukan konsentrasi krim ekstrak kulit buah kopi arabika terbaik terhadap tingkat regenerasi jaringan kulit mencit yang mengalami luka bakar. metode yang digunakan yaitu rancangan acak lengkap (ral), menggunakan 25 ekor mencit berumur 3 bulan dengan rerata berat badan 30 g. luka bakar diameter 1 cm dibuat pada daerah paravertebral. mencit diberi perlakuan berupa k-, k+, k1, k2, dan k3, masing-masing menggunakan basis krim (k-), sulfadiazine (k+), krim ekstrak kulit buah kopi arabika 1% (ki), 3% (k2), dan 5% (k3). pengobatan berupa pengolesan krim sesuai perlakuan sebanyak dua kali sehari selama 14 hari. pembuatan sediaan histologis menggunakan metode parafin dengan pewarnaan hematoxylin eosin. parameter yang dilihat yaitu jumlah pembuluh darah baru, sel fibroblas, folikel rambut, kelenjar minyak, kelenjar keringat dan ketebalan jaringan granulasi. data hasil penelitian dianalisis menggunakan anova, dilanjutkan uji duncan. hasil penelitian menunjukkan bahwa krim ekstrak kulit buah kopi arabika terbukti efektif mempercepat penyembuhan luka bakar pada lapisan dermis kulit mencit ditandai dengan peningkatan neovaskularisasi, perbaikan struktur kulit dan pembentukan jaringan granulasi, di mana konsentrasi 3% optimal pada fase remodeling melalui pembentukan pembuluh darah baru dan penurunan fibroblas, sedangkan konsentrasi 5% menunjukkan efektivitas terbaik secara menyeluruh dengan peningkatan ketebalan jaringan granulasi serta perbaikan struktur jaringan kulit. secara keseluruhan, pemberian krim ekstrak kulit buah kopi arabika mampu meningkatkan proses regenerasi jaringan melalui peningkatan neovaskularisasi, pematangan jaringan granulasi dan perbaikan struktur kulit pada area luka bakar. kata kunci: luka bakar, kulit, krim, kopi arabika



Abstract

This study aimed to determine the effectiveness of ethanol extract cream from the husk of Arabica coffee (Coffea arabica L.) in accelerating the healing of burn wounds in the dermal layer of mice skin and to identify the best concentration of the cream for enhancing skin tissue regeneration in burned mice. The study employed a Completely Randomized Design (CRD), using 25 mice aged 3 months with an average body weight of 30 g. Burn wounds with a diameter of 1 cm were made in the paravertebral area. The mice were treated with five different groups: negative control (K-) using cream base, positive control (K+) using Sulfadiazine and three treatment groups using Arabica coffee husk extract cream at concentrations of 1% (KI), 3% (K2), and 5% (K3). The treatments were applied topically twice daily for 14 days. Histological preparations were made using the paraffin method and stained with Hematoxylin eosin. The observed parameters included the number of newly formed blood vessels, fibroblast cells, hair follicles, sebaceous glands, sweat glands and the thickness of granulation tissue. The research data were analyzed using ANOVA followed by Duncan's test. The results showed that the Arabica Coffe husk extract cream was effective in accelerating burn wound healing in the dermal layer of mice, as indicated by increased neovascularization, improved skin structure, and granulation tissue formation. The 3% concentration was optimal during the remodeling phase through enhanced neovascularization and reduced fibroblast numbers, whereas the 5% concentration demonstrated the best overall effectiveness with increased granulation tissue thickness and improved skin tissue structure. Overall, the application of Arabica coffee husk extract cream enchanced tissue regeneration through increased neovascularization, maturation of granulation tissue, and improved skin structure in the burn wound area.
