Abstrak nama : khaizatul rauza fakultas : kedokteran gigi judul : pengaruh cumi-cumi (loligo sp.) tersulfonasi terhadap pertumbuhan bakteri aggregatibacter actinomycetemcomitans melalui uji khm dan kbm periodontitis merupakan infeksi kronis pada jaringan penyangga gigi dan salah satu bakteri yang berperan penting adalah aggregatibacter actinomycetemcomitans. tulang cumi-cumi (loligo sp.) merupakan sumber kitosan tipe β yang berpotensi dimodifikasi melalui proses sulfonasi untuk meningkatkan aktivitas antibakterinya. penelitian ini bertujuan mengevaluasi kemampuan kitosan tersulfonasi dari tulang cumi-cumi dalam menghambat dan membunuh a. actinomycetemcomitans melalui penentuan konsentrasi hambat minimum (khm) dan konsentrasi bunuh minimum (kbm). kitosan diekstraksi melalui tahap deproteinasi, demineralisasi, dan deasetilasi, kemudian dimodifikasi menggunakan asam sulfat (h₂so₄). uji antibakteri dilakukan dengan metode dilusi dan dianalisis menggunakan uji kruskal-wallis. hasil penelitian menunjukkan bahwa nilai khm diperoleh pada konsentrasi 12,5% dengan nilai od 0,198. nilai kbm ditentukan pada konsentrasi 100% yang ditandai dengan tidak adanya pertumbuhan koloni bakteri. analisis statistik menunjukkan perbedaan signifikan antarperlakuan (p = 0,002). uji lanjutan menunjukkan bahwa pada khm, perbedaan signifikan hanya terjadi antara konsentrasi 12,5% dan kontrol negatif. pada kbm, perbedaan signifikan ditemukan antara konsentrasi 100% dan kontrol positif dibandingkan kontrol negatif. penelitian ini menunjukkan bahwa kitosan tersulfonasi dari tulang cumi-cumi memiliki aktivitas antibakteri yang kuat dan berpotensi dikembangkan sebagai agen antibakteri alami terhadap a. actinomycetemcomitans. kata kunci: kitosan, loligo sp., sulfonasi, aggregatibacter actinomycetemcomit -ans, khm, kbm.



Abstract

Name : Khaizatul Rauza Faculty : Faculty of Dentistry Title : The Effect of Sulfonated Squid Chitosan (Loligo Sp.) on the Growth of Aggregatibacter actinomycetemcomitans through MIC and MBC Tests Periodontitis is a chronic infection of the tooth-supporting tissues, with Aggregatibacter actinomycetemcomitans recognized as a key periodontal pathogen. Squid bone (Loligo sp.) is a potential source of β-type chitosan, which can be chemically modified through sulfonation to enhance its antibacterial properties. This study aimed to evaluate the antibacterial efficacy of sulfonated chitosan derived from squid bone against A. actinomycetemcomitans by determining the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC). Chitosan was extracted via deproteinization, demineralization, and deacetylation processes, followed by sulfonation using sulfuric acid (H₂SO₄). Antibacterial activity was assessed using the broth dilution method, and the data were analyzed using the Kruskal–Wallis test. The results indicated that the MIC was observed at a concentration of 12.5%, with an optical density (OD) value of 0.198. The MBC was achieved at a concentration of 100%, as evidenced by the absence of bacterial growth. Statistical analysis revealed significant differences among treatment groups (p = 0.002). Post hoc analysis showed that at the MIC level, a significant difference was observed only between the 12.5% concentration and the negative control, whereas at the MBC level, significant differences were found between the 100% concentration and both the positive and negative controls. These findings suggest that sulfonated chitosan derived from squid bone exhibits strong antibacterial activity and has potential as a natural antibacterial agent against A. actinomycetemcomitans.
