USRATUL AMNA, UJI AKTIVITAS ANTI-AGING DAN FORMULASI KRIM ANTI-AGING DARI EKSTRAK AKTIF BUNGA JOHAR (CASSIA SIAMEA L.). Banda Aceh MIPA-KIMIA,2025

penelitian ini dilakukan untuk mengevaluasi aktivitas anti-aging serta merumuskan sediaan krim dari ekstrak bunga johar (cassia siamea l.) yang diperoleh menggunakan pelarut metanol, etil asetat, dan n-heksan. proses ekstraksi dilakukan melalui metode maserasi dengan pelarut metanol, kemudian diikuti dengan partisi cair-cair untuk memperoleh ekstrak berdasarkan tingkat kepolarannya. kandungan senyawa metabolit sekunder seperti alkaloid, saponin, flavonoid, fenolik, steroid, terpenoid, dan tanin berhasil diidentifikasi melalui uji fitokimia. aktivitas anti-aging dievaluasi secara in-vitro dengan mengukur penghambatan enzim elastase pankreas babi (porcine pancreatic elastase/ppe), dan asam oleanolat digunakan sebagai kontrol positif. dari hasil pengujian, ditemukan bahwa ekstrak metanol kasar menunjukkan aktivitas penghambatan elastase tertinggi dengan persen inhibisi sebesar 97,53% dan nilai ic₅₀ sebesar 13,895 µg/ml. aktivitas ini diikuti oleh ekstrak metanol partisi (ic₅₀ 22,292 µg/ml), etil asetat (ic₅₀ 24,931 µg/ml), dan n-heksan (ic₅₀ 38,597 µg/ml). sediaan krim yang diformulasikan dari ekstrak metanol kasar menunjukkan karakteristik fisik yang sesuai dengan standar sni-2016-4399-1996, mencakup nilai ph, daya sebar, daya lekat, homogenitas, dan viskositas. formula terbaik ditunjukkan oleh formula 3 (f3), yang diformulasikan dengan konsentrasi ekstrak tertinggi dan menghasilkan nilai ic₅₀ sebesar 95,488 µg/ml. dengan demikian, dapat disimpulkan bahwa ekstrak metanol kasar dari bunga johar berpotensi digunakan sebagai agen anti-aging alami yang efektif dalam sediaan krim topikal. kata kunci: cassia siamea l., anti-aging, porcine pancreatic elastase/ppe, ic50, formulasi.



Abstract

This research was conducted to evaluate the anti-aging activity and formulate a cream preparation from Johar flower (Cassia siamea L.) extracts using methanol, ethyl acetate, and n-hexane as solvents. Extraction was carried out by maceration using methanol, followed by liquid-liquid partitioning to obtain extracts with different polarities. The presence of secondary metabolites such as alkaloids, saponins, flavonoids, phenolics, steroids, terpenoids, and tannins was identified through phytochemical screening. Anti-aging activity was assessed in vitro by measuring the inhibition of porcine pancreatic elastase (PPE), with oleanolic acid used as a positive control. The results showed that the crude methanol extract exhibited the highest elastase inhibitory activity, with a percentage inhibition of 97.53% and an IC₅₀ value of 13.895 µg/mL. This was followed by the partitioned methanol extract (IC₅₀ 22.292 µg/mL), ethyl acetate extract (IC₅₀ 24.931 µg/mL), and n-hexane extract (IC₅₀ 38.597 µg/mL). The cream formulated from the crude methanol extract was found to meet the physical criteria set by the Indonesian National Standard (SNI-2016-4399-1996), including pH, spreadability, adhesion, homogeneity, and viscosity. The best anti-aging performance was shown by Formula 3 (F3), which contained the highest extract concentration and achieved an IC₅₀ value of 95.488 µg/mL. It can be concluded that the crude methanol extract of Johar flower has promising potential as a natural anti-aging agent in topical cream formulations.
