Penelitian ini bertujuan mengetahui sebaran dan ekspresi protein kontraktil alpha-smooth muscle actin (α-sma) pada usus besar ayam kampung (gallus domesticus) dan itik muskovi (cairina moschata) menggunakan teknik imunohistokimia (ihk) metode avidin-biotin complex peroxidase (abc). sampel yang digunakan berupa jaringan usus besar (sekum, kolon, dan rektum) dari masing-masing tiga ekor ayam kampung dan itik muskovi jantan dewasa. jaringan difiksasi dalam neutral buffered formalin 10%, diproses menjadi preparat histologi, dan dilakukan pewarnaan ihk dengan antibodi spesifik α-sma. ekspresi α-sma diamati berdasarkan terbentuknya warna cokelat pada jaringan dan data dianalisis secara deskriptif dan kuantitatif menggunakan skor intensitas (intensity score/is). hasil penelitian menunjukkan bahwa sebaran α-sma baik pada ayam kampung maupun itik terdapat di tunika mukosa, tunika submukosa, dan tunika muskularis, namun tidak ditemukan pada tunika serosa. ekspresi α-sma pada usus besar ayam kampung lebih tinggi secara signifikan dibandingkan itik muskovi. ketebalan otot polos tunika muskularis pada sekum itik muskovi lebih tebal dibandingkan ayam kampung (p



Abstract

This study aims to determine the distribution and expression of the contractile protein alpha-smooth muscle actin (α-SMA) in the large intestine of native chickens (Gallus domesticus) and muscovy ducks (Cairina moschata) using immunohistochemistry (IHC) using the avidin-biotin complex peroxidase (ABC) method. The samples used were large intestine tissue (cecum, colon, and rectum) from three adult male native chickens and muscovy ducks, respectively. The tissue was fixed in 10% neutral buffered formalin, processed into histological slides, and IHC stained with α-SMA-specific antibodies. α-SMA expression was observed based on the formation of brown color in the tissue and the data were analyzed descriptively and quantitatively using an intensity score (IS). The results showed that the distribution of α-SMA in both native chickens and ducks was found in the tunica mucosa, tunica submucosa, and tunica muscularis, but was not found in the tunica serosa. α-SMA expression in the large intestine of native chickens was significantly higher than that of muscovy ducks.
