Abstrak ikan gabus (channa striata) adalah salah satu jenis ikan air tawar yang memiliki nilai ekonomis dan ekologis tinggi di wilayah asia tenggara, termasuk indonesia. tujuan penelitian ini untuk mengkaji konsentrasi ion-ion na+, k+, ca, cl⁻ dan hco₃⁻ dalam seminal plasma sperma ikan gabus, lalu menyusun formulasi larutan ekstender dan menguji efektivitas ekstender tersebut. metode yang digunakan pada penelitian ini terdiri dari 3 (tiga) tahapan kajian, kajian pertama yaitu eksperimental, kajian kedua menggunakan rancangan acak lengkap (ral) terdiri dari 5 perlakuan dan 4 ulangan, kajian ketiga terdiri dari 6 perlakuan 4 ulangan. kajian pertama adalah menganalisis kandungan atau komposisi ion na+, k+, ca, cl⁻ dan hco₃⁻ dalam seminal plasma sperma ikan gabus, kemudian menyusun formula ekstendernya berdasarkan komposisi tersebut. kajian kedua adalah pengujian efektivitas ekstender tersebut dengan membandingkannya dengan ekstender lain yang telah ada, antara lain larutan glucose-base, ringer, nacl fisiologis, larutan urea dan espig. sedangkan kajian ketiga adalah menguji rasio pengenceran sperma menggunakan jenis ekstender terbaik dari hasil eksperimen tahap b, dengan rasio pengenceran 1:0, 1:10, 1:20, 1:30, 1:40, 1:50 dan 1:60 (sperma: ekstender, v/v). berdasarkan hasil penelitian pada setiap kajian didapatkan nilai cairan semen ikan gabus memiliki komposisi ion na+ sebesar 100,12 ppm, ion k+ 4,36 ppm, ca+ 4,57 ppm, cl⁻ 8,58 ppm dan hco₃⁻ didapatkan nilai sebesar 2 ppm. berdasarkan nilai komposisi ion-ion tersebut telah diformulasikan larutan ekstender semua media kriopreservasi sperma ikan gabus yang beri nama ekstender seminal plasma ikan gabus (espig) dengan komposisi adalah: nacl sebesar 0,254 mg/l, kcl 0,0083 mg/l, cacl 0,012 mg/l, dan nahco3 0,00275 mg/l. hasil uji anova menunjukkan bahwa larutan espig berpengaruh nyata terhadap nilai motilitas, viabilitas, abnormalitas, fertilisasi dan durasi sperma hidup sperma ikan gabus (p



Abstract

ABSTRACT The snakehead fish (Channa striata) is a type of freshwater fish with high economic and ecological value in Southeast Asia, including Indonesia. The objective of this study was to examine the concentrations of Na⁺, K⁺, Ca²⁺, Cl⁻, and HCO₃⁻ ions in the seminal plasma of Channa striata sperm, formulate an extender solution, and test its effectiveness. The methodology used in this study consisted of three (3) stages: First stage experimental analysis of the ionic composition (Na⁺, K⁺, Ca²⁺, Cl⁻, and HCO₃⁻) in the seminal plasma of snakehead fish sperm, followed by formulation of an extender solution based on the ionic composition. Second stage effectiveness testing of the formulated extender using a Completely Randomized Design (CRD) with 5 treatments and 4 replications. This stage compared the new extender with existing ones, including Glucose-Base solution, Ringer's solution, physiological NaCl solution, Urea solution, and the newly developed extender named ESPIG (Extender Seminal Plasma Ikan Gabus). Third stage testing various dilution ratios using the best-performing extender from the second stage. The dilution ratios tested were 1:0, 1:10, 1:20, 1:30, 1:40, 1:50, and 1:60 (Sperm: Extender, v/v). The results of the study revealed that the seminal plasma of snakehead fish contains the following ion concentrations: Na⁺ = 100.12 ppm, K⁺ = 4.36 ppm, Ca²⁺ = 4.57 ppm, Cl⁻ = 8.58 ppm, and HCO₃⁻ = 2 ppm. Based on these values, an extender solution was formulated, named ESPIG (Extender Seminal Plasma Ikan Gabus), with the following composition: NaCl = 0.254 mg/L, KCl = 0.0083 mg/L, CaCl₂ = 0.012 mg/L, and NaHCO₃ = 0.00275 mg/L. NOVA test results showed that ESPIG had a significant effect on motility, viability, abnormality, fertilization, and sperm lifespan of Channa striata sperm (p < 0.05). ESPIG produced the following respective values. ESPIG recorded the following values: motility 58.00%, viability 35.33%, abnormality 0.66%, fertilization 43.33%, and sperm longevity 9.44 minutes. The best dilution ratio for Channa striata sperm was found to be 1:40, with the following results: 53.33% (motility), 43.00% (viability), 1.33% (abnormality), 55.00% (fertilization), and 7.97 minutes (sperm lifespan). Therefore, it can be concluded that the ESPIG solution at a 1:40 dilution ratio is recommended for the dilution of Channa striata sperm.
