Severe early childhood caries (s-ecc) merupakan bentuk progresif dari karies gigi anak usia dini yang berkaitan erat dengan keberadaan streptococcus mutans. penggunaan bahan antibakteri alami seperti daun kopi arabika (coffea arabica) gayo menjadi alternatif yang potensial dalam menghambat pertumbuhan bakteri tersebut. penelitian bertujuan untuk mengetahui pengaruh ekstrak daun kopi arabika (coffea arabica) gayo terhadap pertumbuhan streptococcus mutans dari isolat plak anak dengan s-ecc. penelitian menggunakan desain eksperimental laboratorium dengan rancangan pre test and post test only control group. sampel berupa ekstrak etanol daun kopi arabika (coffea arabica) gayo pada konsentrasi 12,5%, 25%, 50%, dan 100% diuji terhadap isolat s. mutans. kelompok kontrol positif menggunakan klorheksidin 0,2% dan kontrol negatif menggunakan akuades steril. uji dilakukan menggunakan metode dilusi cair dan diukur dengan spektrofotometer uv-vis. h a i l penelitian menunjukkan ekstrak daun kopi arabika (coffea arabica) gayo mampu menghambat pertumbuhan s. mutans secara signifikan. nilai hambat terendah mulai tampak pada konsentrasi 12,5%, menunjukkan pengaruh potensial ekstrak daun kopi arabika (coffea arabica). ekstrak daun kopi arabika (coffea arabica) gayo memiliki potensi sebagai antibakteri alami terhadap s. mutans isolat plak anak s-ecc.



Abstract

Severe Early Childhood Caries (S-ECC is a progressive form of early childhood dental caries that is closely related to the presence of Streptococcus mutans. The use of natural antibacterial ingredients such as Gayo Arabica coffee leaves (Coffea arabica) is a potential alternative in inhibiting the growth of these bacteria. This study aims to determine the Antibacterial Activity of Gayo Arabica coffee leaf extract against the growth of Streptococcus mutans from children's plaque isolates with S-ECC. This study used a laboratory experimental design with a pre test and post test only control group design. Samples of Gayo Arabica coffee leaf ethanol extract at concentrations of 12.5%, 25%, 50%, and 100% were tested against S. mutans. The positive control group used 0.2% chlorhexidine and the negative control used sterile distilled water. Antibacterial Activity test was conducted using dilution method and measured by UV-Vis spectrophotometer. The results showed that Gayo Arabica coffee leaf extract was able to inhibit the growth of S. mutans significantly. The lowest effective concentration was recorded at 12.5%, indicating the potential antibacterial activity of Arabica coffee leaf extract. Gayo Arabica coffee leaf extract has potential as a natural antibacterial against S. mutans isolated from S-ECC.
